Abstract
Spirulina sp. is a microalga rich in proteins, lipids, minerals, and natural pigments such as phycocyanin, β-carotene, and chlorophyll, which possess antioxidant properties. Its biochemical composition is strongly influenced by nutrient availability and cultivation conditions. Walne fertilizer is widely applied in Spirulina cultivation but is relatively expensive, prompting the need for alternative nutrient sources. This study employed a Completely Randomized Design (CRD) with three treatments Walne fertilizer, artificial fertilizer, and a mixture of both each with three replications. The experiment consisted of two phases: cultivation and harvesting, followed by phycocyanin extraction. Results showed that growth rates were generally similar across treatments, although a decline in optical density occurred on day 21 in the artificial and mixed fertilizer treatments. Protein content showed no significant difference, with the highest mean value (6.03%) obtained from the artificial fertilizer treatment. The highest wet biomass yield (0.09%) was observed with Walne fertilizer, while the artificial fertilizer treatment yielded the highest phycocyanin (33.48%). The greatest phycocyanin concentration (2.79%) was recorded in both Walne and artificial fertilizer treatments. The best antioxidant activity was observed in the treatment with mixed fertilizer addition (185.85 μg/mL). These findings indicate that artificial fertilizer is comparable to Walne fertilizer in terms of protein content, biomass yield, phycocyanin yield, and concentration, while Walne fertilizer remains superior in supporting growth rate. In addition, the mixed fertilizer outperformed in terms of phycocyanin yield and antioxidant activity.

| Additional Information: | Spirulina sp. merupakan mikroalga yang kaya akan protein, lemak, mineral, serta pigmen alami seperti fikosianin, β-karoten, dan klorofil yang memiliki sifat antioksidan. Komposisi biokimianya sangat dipengaruhi oleh ketersediaan nutrisi dan kondisi kultivasi. Pupuk Walne banyak digunakan dalam budidaya Spirulina, tetapi harganya relatif mahal sehingga diperlukan sumber nutrisi alternatif. Penelitian ini menggunakan Rancangan Acak Lengkap (RAL) dengan tiga perlakuan, yaitu pupuk Walne, pupuk buatan, dan campuran keduanya, masing- masing dengan tiga ulangan. Penelitian dilaksanakan dalam dua tahap, yakni kultivasi dan pemanenan, kemudian dilanjutkan dengan ekstraksi fikosianin. Hasil penelitian menunjukkan bahwa laju pertumbuhan relatif serupa pada semua perlakuan, meskipun terjadi penurunan nilai optical density pada hari ke-21 pada perlakuan pupuk buatan dan campuran. Hasil yang diperoleh menunjukkan tidak terdapat perbedaan nyata pada parameter protein, rendemen biomassa basah, rendemen fikosianin, konsentrasi fikosianin dan aktivitas antioksidan. Kandungan protein tidak menunjukkan perbedaan yang signifikan, dengan nilai rata-rata tertinggi (6,03%) diperoleh pada perlakuan pupuk buatan. Rendemen biomassa basah tertinggi (0,09%) diperoleh pada perlakuan pupuk Walne, sedangkan rendemen fikosianin tertinggi (33,48%) terdapat pada perlakuan pupuk campuran. Konsentrasi fikosianin tertinggi (2,79%) dicapai pada perlakuan pupuk Walne dan pupuk buatan. Aktivitas antioksidan terbaik berada pada perlakuan penambahan pupuk campuran (185,85μg/mL). Temuan ini menunjukkan bahwa pupuk buatan memiliki efektivitas yang sebanding dengan pupuk Walne dalam hal kadar protein, rendemen biomassa, dan konsentrasi fikosianin. Namun, pupuk Walne tetap lebih unggul dalam mendukung laju pertumbuhan. Serta pupuk campuran unggul dalam rendemen fikosianin dan aktivitas antioksidan. | |||||||||
