Abstract

This study aims to examine endophytic fungi in Zingiberaceae plants on Mount Tukung Gede and implement the findings into a biology practicum module. The research was conducted from July 2022 to February 2023, with root samples collected using an exploratory method at three different stations based on vegetation types. Root isolation was performed on PDA (Potato Dextrose Agar) medium, followed by macroscopic and microscopic observations. Macroscopic identification included colony color, texture, and diameter, while microscopic identification focused on the presence of septa, hyphal pigmentation, and the morphology of conidia and conidiophores. A total of 25 isolates were obtained, consisting of three genera: Aspergillus sp. (13 isolates), Penicillium sp. (2 isolates), and Fusarium sp. (10 isolates). Station III showed the highest number of isolates, likely due to higher humidity levels. The dominant endophytic fungi at Station III included Aspergillus sp2, Fusarium sp3, and Penicillium spp. The research findings were incorporated into a biology practicum module to enhance student understanding. The module is systematically structured from title to discussion materials, supported by color images and pretest/post-test components to strengthen the learning process.

Additional Information: Penelitian ini bertujuan mengkaji cendawan endofit pada tumbuhan Zingiberaceae di Gunung Tukung Gede dan mengimplementasikannya dalam modul praktikum biologi. Penelitian dilaksanakan dari Juli 2022 hingga Februari 2023 dengan pengambilan sampel akar menggunakan metode eksplorasi di tiga stasiun berbeda berdasarkan vegetasi. Isolasi akar dilakukan pada media PDA, lalu diamati secara makroskopis dan mikroskopis. Identifikasi makroskopis mencakup warna koloni, tekstur, dan diameter, sedangkan mikroskopis mencakup septat, pigmentasi hifa, dan bentuk konidia serta konidiofor. Dari 25 isolat yang diperoleh, ditemukan tiga jenis cendawan endofit: Aspergillus sp (13 isolat), Penicillium sp (2 isolat), dan Fusarium sp (10 isolat). Stasiun III menunjukkan jumlah isolat terbanyak, diduga karena kelembapan yang lebih tinggi. Tiga jenis cendawan dominan di stasiun III adalah Aspergillus sp2, Fusarium sp3, dan Penicillium spp. Hasil penelitian diintegrasikan ke dalam modul praktikum biologi untuk memudahkan pemahaman peserta didik. Modul disusun secara sistematis mulai dari judul hingga bahan diskusi, dilengkapi gambar berwarna serta pretest dan post-test untuk menunjang proses belajar.
