Abstract

One of the endemic Alocasia species of Indonesia that was successfully classified by botanical name in 1997 in its native Kalimantan area is Alocasia rugosa or better known as Alocasia Melo Badak. Alocasia rugosa is now almost endangered, because its growth is slow. In vitro propagation of Alocasia rugosa with callus proliferation can produce uniform seedlings simultaneously and quickly using additional growth regulators Thidiazuron (TDZ) and 6-Benzyl Amino Purine (BAP) and a culture system. This study aims to determine and obtain the best media composition and culture system for organogenic callus proliferation of Alocasia rugosa 'Melo Badak'. This research was conducted at the Tissue Culture Laboratory of the Soekarno BRIN Cibinong Science and Technology Area (KST) from October 2024 to January 2025, using a 2 factor Completely Randomized Design (CRD). The first factor was the media composition consisting of 4 levels (M1 = MS + 0.25 mg/l TDZ + 0.5 mg/l BAP, M2 = MS + 0.5 mg/l TDZ + 0.25 mg/l BAP, M3 = ½ MS + 0.25 mg/l TDZ + 0.5 mg/l BAP, M4 = ½ MS + 0.5 mg/l TDZ + 0.25 mg/l BAP) and the second factor was the culture system consisting of 4 levels (K1 = solid, K2= liquid, K3 = static thin film layer, K4 = dynamic thin film layer). The results showed that the composition of M1 media (MS + 0.25 mg/l TDZ + 0.5 mg/l BAP) gave the best results on the proliferation of organogenic callus of Alocasia rugosa ‘Melo Badak’ on the parameters of callus size at 12 Weeks After Planting (WAP) and wet weight of callus at 4 and 8 WAP. The K2 culture system (liquid) gave the best results on the proliferation of organogenic callus of Alocasia rugosa ‘Melo Badak’ on the parameters of callus size at 8 WAP, percentage of new callus at 6, 10 and 12 WAP, percentage of new shoots at 6, 8 and 10 WAP, wet weight at 4, 8 and 12 WAP and increase in wet weight at 8 WAP. There is an interaction between the media composition and culture system on the proliferation of organogenic callus of Alocasia rugosa ‘Melo Badak’ on the parameters of the percentage of new callus at 4 and 8 WAP and the increase in wet weight at 12 WAP.

Alocasia rugosa kini terancam punah, karena pertumbuhannya lambat. Perbanyakan Alocasia rugosa secara in vitro dengan proliferasi kalus dapat menghasilkan bibit seragam secara bersamaan dan cepat. Penelitian ini bertujuan untuk mengetahui dan mendapatkan komposisi media dan sistem kultur terbaik untuk proliferasi kalus organogenik Alocasia rugosa ‘Melo Badak’. Penelitian ini dilaksanakan di Laboratorium Kultur Jaringan Kawasan Sais dan Teknologi (KST) Soekarno BRIN Cibinong mulai bulan Oktober 2024 sampai dengan Januari 2025, menggunakan Rancangan Acak Lengkap (RAL) 2 faktor. Faktor pertama yaitu komposisi media terdiri dari 4 taraf (M1 = MS + 0,25 mg/l TDZ + 0,5 mg/l BAP, M2 = MS + 0,5 mg/l TDZ + 0,25 mg/l BAP, M3 = ½ MS + 0,25 mg/l TDZ + 0,5 mg/l BAP, M4 = ½ MS + 0,5 mg/l TDZ + 0,25 mg/l BAP) dan faktor kedua yaitu sistem kultur terdiri atas 4 taraf (K1 = padat, K2 = cair, K3 = thin film layer statis, K4 = thin film layer dinamis). Hasil penelitian menunjukkan komposisi media M1 (MS + 0,25 mg/l TDZ + 0,5 mg/l BAP) memberikan hasil terbaik terhadap proliferasi kalus organogenik Alocasia rugosa ‘Melo Badak’ pada parameter ukuran kalus umur 12 MST dan bobot basah kalus umur 4 dan 8 MST. Sistem kultur K2 (cair) memberikan hasil terbaik terhadap proliferasi kalus organogenik Alocasia rugosa ‘Melo Badak’ pada parameter ukuran kalus 8 MST, persentase kalus baru umur 6, 10 dan 12 MST, persentase tunas baru umur 6, 8 dan 10 MST, bobot basah umur 4, 8 dan 12 MST serta pertambahan bobot basah umur 8 MST. Terdapat interaksi antara komposisi media dan sistem kultur terhadap proliferasi kalus organogenik Alocasia rugosa ‘Melo Badak’ pada parameter persentase kalus baru umur 4 dan 8 MST serta pertambahan bobot basah umur 12 MST
