Abstract
This study aimed to identify and characterize rhizosphere bacteria isolated from Mimosa pudica that have the ability to inhibit the growth of Fusarium oxysporum f.sp. cubense. The research was conducted from March to May 2025 at the Soil and Agroclimate Laboratory, Department of Agroecotechnology, Faculty of Agriculture, Sultan Ageng Tirtayasa University. This research was used a Completely Randomized Design (CRD) with a non-factorial arrangement and 4 replications, consisting of 6 treatments with rhizosphere bacterial isolates from Mimosa pudica and 1 control treatment. From the bacterial purification process, 24 rhizosphere bacterial isolates were obtained, of which 6 isolates showed antagonistic properties. Based on the percentage of inhibition (after transformation), the highest inhibition rate at 7 Days After Incubation (DAI) was recorded for isolate BRP 4, with a value of 11.67%. Gram staining using 3% KOH and Gram staining technique showed that isolates BRP 1 and BRP 2 were Grampositive, while isolates BRP 3, BRP 4, BRP 5, and BRP 6 were Gram-negative. Based on macroscopic observation of bacterial colony morphology, BRP 1 and BRP 2 resembled the Bacillus genus, while isolates BRP 3, BRP 4, BRP 5, and BRP 6 resembled the Pseudomonas genus. Pathogenicity tests on the 6 bacterial isolates revealed that BRP 1, BRP 3, and BRP 4 were non-pathogenic to plants, whereas BRP 2, BRP 5, and BRP 6 exhibited pathogenic characteristics.

| Additional Information: | Penelitian ini bertujuan untuk mengetahui dan mengidentifikasi bakteri rhizosfer asal putri malu yang memiliki kemampuan dalam menghambat pertumbuhan cendawan Fusarium oxysporum f.sp. cubense. Penelitian ini dilakukan pada bulan Maret-Mei 2025 di Laboratorium Tanah dan Agroklimat, Jurusan Agroekoteknologi, Fakultas Pertanian, Universitas Sultan Ageng Tirtayasa. Penelitian ini menggunakan Rancangan Acak Lengkap (RAL) non faktorial dengan 4 ulangan yang terdiri dari 6 perlakuan isolat bakteri rhizosfer putri malu dan 1 perlakuan kontrol. Dari pemurnian bakteri didapatkan 24 isolat bakteri rhizosfer putri malu dan terdapat 6 isolat yang bersifat antagonis. Berdasarkan persentase daya hambat setelah ditransformasi tertinggi pada 7 Hari Setelah Inkubasi (HSI) diperoleh oleh isolat BRP 4 yaitu sebesar 11.67%. Pada pengujian gram menggunakan KOH 3% dan pewarnaan gram didapatkan isolat BRP 1 dan BRP 2 merupakan bakteri gram positif dan isolat BRP 3, BRP 4, BRP 5, dan BRP 6 merupakan bakteri gram negatif. Berdasarkan pengamatan morfologi koloni bakteri secara makroskopis diketahui BRP 1 dan BRP 2 memiliki kemiripan dengan bakteri genus Bacillus sp., dan isolat BRP 3, BRP 4, BRP 5, serta BRP 6 memiliki kemiripan dengan bakteri genus Pseudomonas sp. Pada uji patogenitas terhadap 6 isolat bakteri, diperoleh bahwa isolat BRP 1, BRP 3, dan BRP 4 merupakan bakteri yang tidak memiliki sifat patogen, sedangkan isolat BRP 2, BRP 5, dan BRP 6 merupakan bakteri yang bersifat patogen pada tanaman | |||||||||
