Abstract

In biological systems, antioxidants have many functions as protection from oxidative damage. Antioxidants can also prevent and repair cell damage caused by free radicals, thus protecting the body from disease. Some commonly used synthetic antioxidants include BHA, BHT and TBHQ. However, their use is limited because they have bad side effects. Therefore, a natural source of antioxidants from nature is needed, such as seagrass (E. acoroides) which has the potential for bioactive compounds that can act as antioxidants. This study aim to determine the effect of different types of solvents on the characterization of the phytochemical profile and antioxidant potential of seagrass extracts (E. acoroides) produced. This study used a completely randomized design of one factor of solvent type consisting of 8 levels (ethanol 96%, 80%, 60%), methanol (100%, 80%, 60%), ethyl acetate, and chloroform. The results showed that different types of solvents had a significant effect on the level of ethanol 60% on the phytochemical profile and antioxidant activity produced. Seagrass extract with 60% ethanol solvent contains phytochemical profiles, namely, phenolics, flavonoids, alkaloids and tannins. The results of the antioxidant activity test, phenolic content, and the highest flavonoid content were found in the 60% ethanol solvent sample compared to other solvents, namely, the IC50 value was ***±** ppm, the total phenolic content was **.**±*.**mgGAE/g, the total flavonoid content was **.**±*.** mg/QE/g extract.

Additional Information: Dalam sistem biologis, antioksidan memiliki banyak fungsi sebagai perlindungan dari kerusakan oksidatif. Antioksidan juga dapat mencegah dan memperbaiki kerusakan sel yang disebabkan oleh radikal bebas, sehingga melindungi tubuh dari penyakit. Beberapa antioksidan sintetik yang umum digunakan antara lain BHA, BHT dan TBHQ. Akan tetapi, penggunaannya terbatas karena memiliki efek samping yang buruk. Oleh karena itu, diperlukan sumber antioksidan alami dari alam, seperti lamun (E. acoroides) yang berpotensi sebagai senyawa bioaktif yang dapat berperan sebagai antioksidan. Penelitian ini bertujuan untuk mengetahui pengaruh jenis pelarut yang berbeda terhadap karakterisasi profil fitokimia dan potensi antioksidan ekstrak lamun (E. acoroides) yang dihasilkan. Penelitian ini menggunakan rancangan acak lengkap satu faktor jenis pelarut yang terdiri dari 8 taraf (etanol 96%, 80%, 60%), metanol (100%, 80%, 60%), etil asetat, dan kloroform. Hasil penelitian menunjukkan bahwa jenis pelarut yang berbeda memberikan pengaruh yang nyata terhadap kadar etanol 60% terhadap profil fitokimia dan aktivitas antioksidan yang dihasilkan. Ekstrak lamun dengan pelarut etanol 60% mengandung profil fitokimia yaitu fenolik, flavonoid, alkaloid dan tanin. Hasil uji aktivitas antioksidan, kadar fenolik, dan kadar flavonoid tertinggi terdapat pada sampel pelarut etanol 60% dibanding pelarut lainnya yaitu nilai IC50 sebesar ***±** ppm, kadar fenolik total sebesar **.**±*.**mgGAE/g, kadar flavonoid total sebesar **.**±*.** mg/QE/g ekstrak.
