Abstract

Rapid seed testing method is required to obtain information on seed quality within a short time. The radicle emergence test and tetrazolium test are some of methods to estimate the seed viability and vigor. The objective of this research was to determine observation time of radicle emergence test and correlate it with other viability and vigor parameters; determine criteria and vigor classification from the topographic pattern of staining of arabica coffee seed embryos on the tetrazolium test and correlate it with other viability and vigor parameters. The parameters for physiological quality testing were the germination, mean rate germination, vigor index, speed of germination, normal germination dry weight, and sprout growth rate. The experimental design of radicle emergence test used Randomized Completely Block Design with factorial treatment design which consisted of two factors and replicated three times. The first factor was varieties which consisted of three levels: S 795, Gayo 1, and Sigarar Utang. The second factor was germination period which consisted of five levels: 168, 192, 216, 240, and 264 hours. The experiment of tetrazolium test was carried out in one factor Randomized Completely Design, replicated nine times. The factor was varieties which consisted of three levels: S 795, Gayo 1, and Sigarar Utang. The best results for radicle emergence test were obtained from the Sigarar Utang variety (98,87%) in 264 hours germination period. There was an interaction between varieties and germination period. The result showed that strong and moderate correlation between radicle emergence and various physiological quality parameters of the seeds. Tetrazolium test on three varieties of Arabica coffee seeds resulted in staining patterns to predict high vigor seeds, low vigor seed, very low vigor seed that would produce abnormal seedling, and dead seeds. The correlation coefficient between criteria of germination test and criteria of tetrazolium test also the correlation coefficient between criteria of tetrazolium test and various parameters of physiological quality of seeds showed moderate, strong, and very strong correlation. Additional Information: Uji pemunculan radikula dan tetrazolium dibutuhkan untuk menguji kualitas benih kopi Arabika dalam waktu yang cepat. Uji pemunculan radikula dan uji tetrazolium merupakan beberapa metode untuk mengestimasi viabilitas dan vigor benih. Penelitian ini bertujuan untuk (1) memperoleh waktu pengamatan yang tepat pada uji pemunculan radikula, serta memperoleh korelasi antara hasil uji pemunculan radikula dengan berbagai parameter viabilitas dan vigor benih, (2) memperoleh kriteria dan klasifikasi vigor dari pola topografi pewarnaan embrio pada uji tetrazolium serta memperoleh korelasi antara hasil uji tetrazolium dengan berbagai parameter viabilitas dan vigor benih. Rancangan percobaan pada pengujian pemunculan radikula adalah Rancangan Acak Kelompok Faktorial yang terdiri dari dua faktor. Faktor pertama adalah varietas benih, terdiri dari tiga taraf yaitu varietas S 795, Gayo 1, dan Sigarar Utang. Faktor kedua adalah periode pengecambahan, terdiri dari lima taraf yaitu 168, 192, 216, 240, dan 264 jam. Pengujian tetrazolium menggunakan Rancangan Acak Lengkap dengan satu faktor yaitu varietas benih yang terdiri dari tiga taraf yaitu varietas S 795, Gayo 1, dan Sigarar Utang. Hasil penelitian uji pemunculan radikula menunjukkan bahwa hasil terbaik terhadap pemunculan radikula diperoleh dari varietas Sigarar Utang (98,87%) pada periode pengecambahan selama 264 jam dimana terdapat interaksi antara varietas dan periode pengecambahan. Terdapat korelasi yang kuat dan cukup kuat antara hasil uji pemunculan radikula dengan berbagai parameter mutu fisiologis benih. Pengujian tetrazolium pada tiga varietas benih kopi Arabika menghasilkan pola pewarnaan untuk memprediksi kecambah normal kuat, normal lemah, abnormal, dan benih mati. Koefisien korelasi antara kriteria kecambah hasil uji daya berkecambah dengan kriteria kecambah hasil uji tetrazolium serta koefisien korelasi antara hasil uji tetrazolium dengan berbagai parameter mutu fisiologis benih menunjukkan korelasi cukup kuat, kuat, dan sangat kuat.
