Abstract

This research was aimed to know the effect of several concentrations of coconut water and glutamine to multiplication embriosomatis Phalaenopsis amabilis Clones KD-69 in vitro. Research was conducted at the Laboratory of Tissue Culture Ornamental Crops Research Institute (Balithi) in November 2012 until January 2013. Research used Randomized Completely Design (RCD) with two factors. The first factor was the use of coconut water with 5 concentrations, namely 0 ml.l -1 (A4). The second factor was the use of glutamine with 7 degree of concentration, namely 0 mg.l -1 (A0), 50 ml.l -1 (A1), 100 ml.l -1 (A2), 150 ml.l -1 (A3) , 200 ml.l -1 (G4), 125mg.l -1 (G0), 25 mg.l -1 (G1), 50 mg.l -1 (G2), 75 mg.l -1 (G3) , 100 mg.l -1 (G6). So there were 35 combinations and repeated 3 times, so there were 105 units of the experiment totally. The results showed that treatment of coconut water can stimulate the multiplication of the number of embryo per explant at the age of 8 MST (35.57). Treatment of coconut water with a concentration of 50 ml/l (A1) gives the best results to the multiplication of Phalaenopsis amabilis embriosomatis. Glutamine treatment could stimulate the multiplication of the number of embryo per explant at the age of 8 MST (37,6) and 9 MST (47,7). Glutamine treatment with a concentration of 50 mg/l (G2) give the best results to the multiplication of the number of embryo per explant, the average increase in the embryo, and the total embryo produced in vitro. There was no interaction between the concentration of coconut water and glutamine to the multiplication number of embryo per explant in vitro.

Additional Information: Penelitian ini bertujuan untuk mengetahui pengaruh beberapa konsentrasi air kelapa dan glutamin terhadap perbanyakan embriosomatis anggrek Phalaenopsis amabilis Klon KD-69 secara in vitro. Penelitian dilaksanakan di Laboratorium Kultur Jaringan Balai Penelitian Tanaman Hias (Balithi) pada bulan November 2012 sampai Januari 2013. Penelitian menggunakan RAL dengan dua faktor. Faktor pertama adalah penggunaan air kelapa dengan 5 konsentrasi, yaitu 0 ml.l (A0), 50 ml.l -1 -1 (A4). Faktor kedua adalah penggunaan glutamin dengan 7 taraf konsentrasi, yaitu 0 mg.l -1 (A1), 100 ml.l -1 (A2), 150 ml.l 1 (A3), 200 ml.l- (G0), 25 mg.l -1 -1 (G5), dan 150 mg.l -1 (G1), 50 mg.l -1 (G2), 75 mg.l -1 (G3), 100 mg.l -1 (G4), 125 mg.l (G6). Sehingga terdapat 35 kombinasi dan diulang 3 kali, sehingga terdapat 105 satuan percobaan. Hasil penelitian menunjukkan bahwa Perlakuan air kelapa dapat memacu perbanyakan jumlah embrio per eksplan pada umur 8 MST (35,57). Perlakuan air kelapa dengan konsentrasi 50 ml/l (A1) memberikan hasil terbaik terhadap perbanyakan embriosomatis anggrek Phalaenopsis amabilis. Perlakuan glutamin dapat memacu perbanyakan jumlah embrio per eksplan pada umur 8 MST (37,6) dan 9 MST (47,7). Perlakuan glutamin dengan konsentrasi 50 mg/l (G2) memberikan hasil terbaik terhadap perbanyakan jumlah embrio per eksplan, rata-rata pertambahan embrio, dan total embrio yang dihasilkan secara in vitro. Tidak terdapat interaksi antara konsentrasi air kelapa dan glutamin terhadap perbanyakan jumlah embrio per eksplan secara in vitro.
