Abstract

Genetic Analysis of 35 Accession Oil Palm (Elaeis guineensis Jacq.) Origin Cameroon by Microsatellite markers. Muhammad Rizky Aliansyah. The diversity of germplasm collections of national palm oil is still low and the required enrichment of the collection through the exploration of the country to the center of diversity and Germplasm collection of palm oil in Indonesia spread across several research institutions and seed producers. 13 Exploration Company in 2009 acquired 103 accessions collected in the National Gardens SDG Sawit. This study uses 35 accessions of crude palm oil palm election was 5 years old has been with the criteria of the average weight of 4 to 9.5 above pertandan young fruit Kilogram.Tujuan this study was to analyze 35 accessions of oil origin the National Gardens SDG Oil Sijunjung, Sumatra west to: (1) determine the level of polymorphism SSR markers; (2) determine the kinship between accession; and (3) analyze aksesiaksesi potential for germplasm collection. Genetic material used leaves from the 35 accession protocol and the DNA is isolated mnggunakan Plant Genomic DNA Mini Kit (Geneaid). DNA was analyzed using 36 SSR markers. Kinship dendrogram constructed using Unweighted Pair-Group Method arithmetic (UPGMA) through the Numerical Taxonomy and Multivariate System (NTSYS) version 2.1-pc. SSR markers with 36 markers were tested on 35 accessions of oil Cameroon produces a total of 207 alleles with the average number of alleles per locus ranged from 1-12 5.75 alleles per locus SSR. The level of polymorphism SSR markers tested is low with the average value of PIC 0.36 range at 0.01 to 0.91. Testing Filogenik diversity divide on 4 groups based on diversity coefficient (KK). The first group consisted of 22 accessions KK 0.70 to 0.77 with a level of diversity of 23-33%. The second group consists of 3 accession KK 0.67 to 0.69 with a level of diversity of 21-23%. The third group consists of 6 accession KK, 0.67 to 0.74 with a level of diversity 26-33% and the fourth group consisted of 4 accessions KK 0.69 to 0.76 with a level of 24-31% .Aksesi diversity with high genetic diversity and that are in different clusters potentially be used as a prospective parent in oil palm breeding program.

Materi genetik yang digunakan daun dari 35 aksesi kemudian DNA diisolasi mnggunakan protokol Genomic DNA Mini Kit Plant (Geneaid) . DNA dianalisis menggunakan 36 marka SSR. Dendrogram kekerabatan dikonstruksi menggunakan Unweighted Pair-Group Method Arithmatic (UPGMA) melalui program Numerical Taxonomy and Multivariate System (NTSYS) versi 2.1-pc. Marka SSR dengan 36 marka yang diuji pada 35 aksesi sawit Kamerun menghasilkan alel total sebanyak 207 dengan rataan jumlah alel per lokus 5,75 berkisar antara 1-12 alel per lokus SSR. Tingkat polimorfisme marka SSR yang diuji tergolong rendah dengan rataan nilai PIC 0,36 berkisar pada 0,01 sampai 0,91. Pengujian Filogenik membagi diversitas atas 4 kelompok berdasar Koefisien Keragaman (KK). Kelompok pertama terdiri atas 22 aksesi KK 0,70 - 0,77 dengan tingkat keragaman 23-33%. Kelompok kedua terdiri atas 3 aksesi KK 0,67-0,69 dengan tingkat keragaman 21-23%. Kelompok ketiga terdiri dari 6 aksesi KK ,0,67-0,74 dengan tingkat keragaman 26-33% dan kelompok keempat terdiri dari 4 aksesi KK 0,69-0,76 dengan tingkat keragaman 24-31%.Terdapat beberapa seperti aksesi tunggal D91(10) kelompok I dengan KK gentik 67,5% dan D20 (7) kelompok III dengan koefisien kesamaan genetik 67% pada dendogram bisa dikatakan berdiri sendiri terhadap aksesi lainnya. Aksesi dengan diversitas genetik tinggi dan berada pada klaster berbeda potensial digunakan sebagai calon tetua dalam program pemuliaan kelapa sawit.
