Koch) possess great potential to be developed as a functional food ingredient, although their utilization remains limited. Phytochemical compounds in beneng taro leaves can provide health benefits but may also cause toxic effects when consumed excessively. Therefore, this study aimed to evaluate the acute toxicity level and phytochemical content of the methanol extract of beneng taro leaves as a basis for determining the safe consumption limit in food-based applications. The research was conducted experimentally using a non-factorial Randomized Block Design (RBD) with one factor, namely the concentration of methanol extract of beneng taro leaves, consisting of six treatment levels (10, 20, 40, 80, 160, and 320 ppm). The acute toxicity test was carried out for 96 hours using zebrafish embryos (Danio rerio) to determine the Lethal Concentration 50 (LC50) value through probit analysis, followed by an Analysis of Variance (ANOVA). The results showed that the LC₅₀ value of 51.02 ppm was classified as slightly toxic. In addition, beneng taro leaf methanol extract had a significant effect on the hatchability and mortality of zebrafish embryos, but had no significant effect on the heart rate and abnormalities of zebrafish embryos. This concentration still causes physiological disturbances, as indicated by a decrease in hatchability and an increase in mortality, suggesting a toxic response of the embryos to the secondary metabolites in the extract.
