Panama disease is a Fusarium wilt disease caused by Fusarium oxysporum f. sp. cubense (Foc). Tropical Race 4 (TR4) is the most virulent type of Foc that poses a serious threat to banana cultivation worldwide. Until now, there is no effective method for handling Tropical Race 4. This research aims to study the potential of endophytes as phytovaccines against Panama disease. The methods used were optimization of growing media for endophytic Fusarium on potato, red bean, and mung bean media and infection of endophytes on Cavendish varieties to induce resistance. Four Fusarium endophyte isolates were tested on two banana varieties, one resistant to TR4 using Rejang and one susceptible using Cavendish. Endophytic Fusarium inoculation and Tropical Race 4 infection were carried out using the simultaneous pouring method. Results showed that potato media produced the highest conidia and isolate NMC 258 had the greatest potential to produce conidia. The four endophytic Fusarium isolates NMC 250, NMC 258, NMC 266, and NMC 267 were able to reduce disease symptoms in Cavendish bananas and did not trigger disease symptoms in Rejang bananas. The PCR amplification results show clear and bright DNA bands but there are faint DNA bands. This study shows that endophytic Fusarium has the potential as an environmentally friendly phytovaccine agent and is recommended as a learning resource for practicum modules in plant growth and development material in the Merdeka Curriculum for class XI SMA.
