%L eprintuntirta12510 %A ILHAM BINTANG %T Pengaruh Konsentrasi Kinetin dan GA (Giberelin Acid) Terhadap Inisiasi Tunas Serta Konsentrasi 2,4-D (2,4-Diklorofenoksiasetat) dan TDZ (Thidiazuron) terhadap Inisiasi Kalus Pada Anggrek Bulan (Phalaenopsis amabilis) Melalui Tangkai Bunga. %D 2016 %X This research was aimed to obtain the effect of hormone concentrations on bud and callus initiation by using flower stalk explant of Phalaenopsis amabilis. This research has been concucted on August until December 2014 at Tissue Culture laboratorium, Indonesian Center for Agricultural Biotechnology, Genetic Resources Research and Development, Bogor, Indonesia (ICABGRRD). Experiment was arranged in factorial by using completely randomized design (CRD). Base medium used is Murashige dan Skoog (1962) (MS), this experiment consists of bud and callus initiation. Shoot initiation from flower stalk consisted of two factors, first factor was Kinetin (0 and 3 mg/l) and the second factor was Geberelic Acid (GA ) (0, 3 and 5 mg/l) with three replication. So its consists 18 experimental unit. Callus initiation from in vitro leaf consisted of one factor with three levels of 2,4-D (0, 2 and 4 mg/l) and three levels of thidiazuron (0, 0.3 and 0.5 mg/l), each replication three times. So its consists 18 experimental unit. There was significant interaction beetwen Kinetin and GA 3 to the shoot initiation. The best modofication for Shoot initiation are kinetin 0 mg/l and GA v 3 ) 2,34 DAP on bud growth and Kinetin 3 mg/l and GA 3 5 mg/l (K ) 0,89 cm on plant high. There was no significant interaction between 2,4D and thidiazuron for callus initiation on all parameter.
