Abstract

The leaves of beneng taro (Xanthosoma undipes K. Koch) possess great potential to be developed as a functional food ingredient, although their utilization remains limited. Phytochemical compounds in beneng taro leaves can provide health benefits but may also cause toxic effects when consumed excessively. Therefore, this study aimed to evaluate the acute toxicity level and phytochemical content of the methanol extract of beneng taro leaves as a basis for determining the safe consumption limit in food-based applications. The research was conducted experimentally using a non-factorial Randomized Block Design (RBD) with one factor, namely the concentration of methanol extract of beneng taro leaves, consisting of six treatment levels (10, 20, 40, 80, 160, and 320 ppm). The acute toxicity test was carried out for 96 hours using zebrafish embryos (Danio rerio) to determine the Lethal Concentration 50 (LC50) value through probit analysis, followed by an Analysis of Variance (ANOVA). The results showed that the LC₅₀ value of 51.02 ppm was classified as slightly toxic. In addition, beneng taro leaf methanol extract had a significant effect on the hatchability and mortality of zebrafish embryos, but had no significant effect on the heart rate and abnormalities of zebrafish embryos. This concentration still causes physiological disturbances, as indicated by a decrease in hatchability and an increase in mortality, suggesting a toxic response of the embryos to the secondary metabolites in the extract. Phytochemical analysis revealed that the methanol extract contained phenolic compounds (14.75 mg GAE/g), flavonoids (64.64 mg QE/g), tannins (16.09 mg TAE/g), saponins (0.89 mg GAE/g), and alkaloids (28.80 mg KE/g).

Additional Information: Daun talas beneng (Xanthosoma undipes K. Koch) memiliki potensi besar untuk dikembangkan sebagai bahan pangan fungsional, meskipun pemanfaatannya masih terbatas. Senyawa fitokimia yang terkandung dalam daun talas beneng dapat memberikan manfaat kesehatan, tetapi juga berpotensi menimbulkan efek toksik apabila dikonsumsi secara berlebihan. Oleh karena itu, penelitian ini bertujuan untuk mengevaluasi tingkat toksisitas akut dan kandungan fitokimia ekstrak metanol daun talas beneng sebagai dasar dalam menentukan batas aman konsumsi pada aplikasi pangan. Penelitian ini dilakukan secara eksperimental menggunakan Rancangan Acak Kelompok (RAK) non faktorial dengan satu faktor, yaitu konsentrasi ekstrak metanol daun talas beneng, yang terdiri dari enam taraf perlakuan (10, 20, 40, 80, 160, dan 320 ppm). Uji toksisitas akut dilakukan selama 96 jam menggunakan embrio ikan zebra (Danio rerio) untuk menentukan nilai Lethal Concentration 50 (LC₅₀) melalui analisis probit, yang dilanjutkan dengan Analisis Varian (ANOVA). Hasil penelitian menunjukkan bahwa nilai LC₅₀ sebesar 51,02 ppm tergolong dalam kategori sedikit toksik. Selain itu, ekstrak metanol daun talas beneng memberikan pengaruh signifikan terhadap daya tetas dan mortalitas embrio ikan zebra, namun tidak memberikan pengaruh signifikan terhadap detak jantung dan abnormalitas embrio ikan zebra. Konsentrasi tersebut masih menyebabkan gangguan fisiologis, yang ditunjukkan oleh penurunan daya tetas dan peningkatan mortalitas, sehingga mengindikasikan adanya respons toksik embrio terhadap metabolit sekunder dalam ekstrak. Analisis fitokimia menunjukkan bahwa ekstrak metanol mengandung senyawa fenolik (14,75 mg GAE/g), flavonoid (64,64 mg QE/g), tanin (16,09 mg TAE/g), saponin (0,89 mg GAE/g), dan alkaloid (28,80 mg KE/g).
