Abstract

Pineapple (Ananas comosus) is a tropical fruit that grows in Indonesia, with a total production of 3.2 million tons in 2022. The most widely cultivated varieties are Smooth Cayenne and Queen. A significant portion of the pineapple, estimated at 55 – 70%, is non-consumable and categorized as waste, which includes the pith, skin, leaves, stems, and crown. This pineapple waste is a rich source of the enzyme bromelain, a protease with high potential for utilization in various industries, particularly the food sector. The purpose of this study was to determine the effect of different types of waste, pineapple varieties, and the interaction between these two factors on the characteristics of the liquid bromelain enzyme solution, including protein content, enzyme activity, specific enzyme activity, and enzyme yield. This was an experimental study conducted using a two-factor factorial randomized design. The first factor was the type of waste, consisting of pineapple flesh, pith, skin, stems, and crown. The second factor was the pineapple variety, consisting of the Smooth Cayenne and Queen varieties. The results indicate that the type of pineapple waste factor and the interaction between the two factors had a highly significant effect on all measured responses. Conversely, the pineapple variety factor only significantly affected enzyme activity and specific enzyme activity. The highest specific enzyme activity values were obtained from the Smooth Cayenne stem (0,620 U/mg) and crown (0,600 U/mg) samples. The largest yields were recorded in the Smooth Cayenne skin (0,185%), Queen crown (0,185%), and Queen leaves (0,185%) samples.

Additional Information: Nanas (Ananas comosus) adalah buah tropis yang tumbuh di Indonesia, dengan total produksi sebesar 3,2 juta ton pada tahun 2022. Varietas yang paling banyak dibudidayakan adalah Smooth Cayenne dan Queen. Sebagian besar bagian nanas, diperkirakan sebesar 55 – 70%, tidak bisa dikonsumsi dan dikategorikan sebagai limbah, yang mencakup bonggol, kulit, daun, batang, dan mahkota. Limbah nanas ini merupakan sumber enzim bromelain yang kaya, yaitu protease dengan potensi tinggi untuk dimanfaatkan di berbagai industri, khususnya sektor pangan. Tujuan penelitian ini adalah untuk mengetahui pengaruh jenis limbah yang berbeda, varietas nanas, dan interaksi antara kedua faktor tersebut terhadap karakteristik larutan enzim bromelain cair, termasuk kandungan protein, aktivitas enzim, aktivitas spesifik enzim, dan hasil enzim. Penelitian ini merupakan penelitian eksperimental yang dilakukan dengan menggunakan rancangan acak lengkap faktorial dua faktor. Faktor pertama adalah jenis limbah, yang terdiri dari daging nanas, bonggol, kulit, batang, dan mahkota. Faktor kedua adalah varietas nanas, yang terdiri dari varietas Smooth Cayenne dan Queen. Hasil menunjukkan bahwa jenis limbah nanas dan interaksi antara kedua faktor tersebut berpengaruh sangat signifikan terhadap semua respons yang diukur. Sebaliknya, faktor varietas nanas hanya berpengaruh signifikan terhadap aktivitas enzim dan aktivitas enzim spesifik. Nilai aktivitas enzim spesifik tertinggi diperoleh dari batang Smooth Cayenne (0,620 U/mg) dan sampel mahkota (0,600 U/mg). Hasil tertinggi tercatat pada kulit Smooth Cayenne (0,185%), mahkota Queen (0,185%), dan daun Queen (0,185%).
