Abstract
This study aims to determine the growth of orchid explants Grammatophyllum speciosum Blume on anorganic fertilizer media and the concentration of 6-Benzylaminopurine (BAP) in vitro. This research was experimental research conducted from December 2024 to March 2025 at the Laboratory of Plant Physiology & Biotechnology, Faculty of Agroecotechnology, Sultan Ageng Tirtayasa University. This study used a Randomized Blocks Design (RBD) consisting of 2 treatment factors. The first factor was the concentration level of anorganic fertilizer which consists of 4 levels, namely: P0 = 0 g/l, P1 = 1 g/l, P2 = 2 g/l, P3 = 3 g/l. The second factor was the concentration level of 6-Benzylaminopurine (BAP) which consists of 4 levels, namely: B0 = 0 ppm, B1 = 1 ppm, B2 = 2 ppm, B3 = 3 ppm. The observation parameters observed in this study were shoot emergence time (WAP), number of buds (piece), number of leaves (strands), explant height (cm), percentage of contaminated explants (%), percentage of browning explants (%) and explant color. The results showed that the concentration level of anorganic fertilizer significantly affected the number of leaves, namely 5.16 at 10 Week After Plant (WAP) and 5.75 at 12 Week After Plant (WAP). Meanwhile, the 6-Benzylaminopurine factor had no significant effect on all parameters and there was an interaction between the concentration level of 6-Benzylaminopurine and anorganic fertilizer.

| Additional Information: | Penelitian ini bertujuan untuk mengetahui pertumbuhan eksplan anggrek Grammatophyllum speciosum Blume pada media pupuk anorganik dan konsentrasi 6-Benzylaminopurine (BAP) secara in vitro. Penelitian ini merupakan penelitian eksperimen yang dilaksanakan pada bulan Desember 2024 sampai dengan Maret 2025 di Laboratorium Fisiologi & Bioteknologi Tanaman, Jurusan Agroekoteknologi, Universitas Sultan Ageng Tirtayasa. Penelitian ini menggunakan Rancangan Acak Kelompok (RAK) yang terdiri dari 2 faktor perlakuan. Faktor pertama adalah konsentrasi pupuk anorganik yang terdiri dari 4 taraf, yaitu: P0 = 0 g/l, P1 = 1 g/l, P2 = 2 g/l, P3 = 3 g/l. Faktor kedua adalah konsentrasi 6-Benzylaminopurine (BAP) yang terdiri dari 4 taraf, yaitu: B0 = 0 ppm, B1 = 1 ppm, B2 = 2 ppm, B3 = 3 ppm. Parameter pengamatan yang diamati dalam penelitian ini adalah waktu munculnya tunas (MST), jumlah tunas (buah), jumlah daun (helai), tinggi eksplan (cm), persentase eksplan terkontaminasi (%), persentase eksplan yang berwarna kecoklatan (%), dan warna eksplan. Hasil penelitian menunjukkan bahwa konsentrasi pupuk anorganik berpengaruh nyata terhadap jumlah daun, yaitu 5,16 pada 10 MST dan 5,75 pada 12 MST. Sementara itu, faktor 6-Benzylaminopurine tidak berpengaruh nyata terhadap semua parameter dan terdapat interaksi antara konsentrasi 6-Benzylaminopurine dan pupuk anorganik. | |||||||||
