Abstract

Acne (Acne vulgaris) is a skin disorder characterized by inflammation due to blockage of the sebaceous gland ducts, which is then exacerbated by bacterial activity, causing an inflammatory process. The use of natural ingredients such as patchouli oil can accelerate acne healing because patchouli oil has potential as an anti-inflammatory. Patchouli oil is applied in the cosmetics, pharmaceutical, and fragrance industries. This study aims to determine the effect of microwave power and extraction time, identify the active compounds of patchouli oil produced, test the ability and analyze the anti-inflammatory potential of patchouli oil using the in silico/molecular docking method, and obtain antibacterial inhibition in the resulting patchouli oil. This study began with drying, grinding, and sieving patchouli leaves, then the extraction process was carried out using the Microwave-Assisted Hydrodistillation (MAHD) method with variations in microwave power of 200, 400, and 600 Watts, and variations in extraction time for 30, 60, 90, and 120 minutes. The results showed that 400 Watt power with 90 minutes gave the highest yield of 5.20%. The results showed that 400 Watt power with 90 minutes gave the highest yield of 5.20%. The resulting patchouli oil contains bioactive compounds patchouli alcohol, δ-Guaiene, β-Caryophyllene, as well as groups of compounds such as flavonoids, alkaloids, and terpenoids. Patchouli oil has the potential as an anti-inflammatory because of bioactive compounds such as α-Copaene, δ-Guaiene, and Alloaromadendrene which have been proven by the molecular docking method and also have antibacterial activity on Staphylococcus aureus bacteria which is able to inhibit 52.5% and Propionibacterium acnes bacteria which is able to inhibit 46.8%.

Additional Information: Jerawat (Acne vulgaris) adalah kelainan kulit yang ditandai oleh timbulnya peradangan akibat tersumbatnya saluran kelenjar sebaceous, yang kemudian diperparah oleh aktivitas bakteri sehingga menyebabkan proses inflamasi. Penggunaan bahan alami seperti minyak nilam (patchouli oil) dapat mempercepat penyembuhan jerawat karena patchouli oil memiliki potensi sebagai antiinflamasi. Patchouli oil diaplikasikan dalam industri kosmetik, farmasi, wewangian. Penelitian ini bertujuan untuk mengetahui pengaruh daya microwave dan waktu ekstraksi, mengidentifikasi senyawa aktif patchouli oil yang dihasilkan, menguji kemampuan dan menganalisis potensi antiinflamasi pada patchouli oil dengan metode in silico/molecular docking, serta mendapatkan penghambatan antibakteri pada patchouli oil yang dihasilkan. Penelitian ini diawali dengan pengeringan, penghalusan dan pengayakan daun nilam, kemudian dilakukan proses ekstraksi dengan metode Microwave-Assisted Hydrodistillation (MAHD) dengan variasi daya microwave 200, 400, dan 600 Watt, serta variasi waktu ekstraksi selama 30, 60, 90 dan 120 menit. Hasil menunjukkan bahwa daya 400 Watt dengan waktu 90 menit memberikan yield tertinggi yaitu 5,20%. Patchouli oil yang dihasilkan mengandung senyawa bioaktif patchouli alcohol, δ-Guaiene, β-Caryophyllene, serta golongan senyawa seperti flavonoid, alkaloid, dan terpenoid. Patchouli oil memiliki potensi sebagai antiinflamasi karena senyawa bioaktif seperti α-Copaene, δ-Guaiene, dan Alloaromadendrene yang telah dibuktikan dengan metode molecular docking dan juga memiliki aktivitas antibakteri pada bakteri Staphylococcus aureus yang mampu menghambat sebesar 52,5% dan bakteri Propionibacterium acnes yang mampu menghambat 46,8%.
