Abstract
The utilization of high concentrations of alkali during the extraction of carrageenan was a subject of concern due to its potential adverse environmental and health impacts. The incorporation of cellulase enzymes serves to reduce the necessity of alkali and accelerate the extraction process. The aim of this research was to determine the optimum conditions for the extraction process of carrageenan from the seaweed Kappaphycus alvarezii with treatments of cellulase enzyme concentration, calcium hydroxide (Ca(OH)2) concentration, and alkali soaking temperature using Response Surface Methodology (RSM). The present study employed the Box-Behnken RSM on the Design Expert 13.0.5.0 program, incorporating three research variables. The variables employed in the experimental design included the cellulase enzyme concentration (0, 1; 0,55;and 1%), calcium hydroxide (Ca(OH)2) concentration (2, 5, and 8%), and alkali soaking temperature (50, 60, and 70°C). The results of the Box-Behnken RSM analysis indicated that the optimum formula was obtained with a desirability value of 0.89, which corresponds to an optimization target of 89%. The optimal formula, as determined by this method, involves the use of a Ca(OH)2 concentration of 2%, a cellulase enzyme concentration of 0.6%, and an alkali soaking temperature of 70°C. The resulting characteristics are as follows: a yield value of 3,951%, gel strength of 931,978 g/cm², viscosity of 198 cP, molecular weight of 172,612 kDa, moisture content of 11,936%, and ash content of 18,027%.

| Additional Information: | Penggunaan alkali dengan konsentrasi tinggi dalam proses ekstraksi karaginan menjadi perhatian karena berpotensi menimbulkan dampak negatif terhadap lingkungan dan kesehatan. Penambahan enzim selulase dapat mengurangi kebutuhan penggunaan alkali serta mempercepat proses ekstraksi. Penelitian ini bertujuan untuk menentukan kondisi optimum dalam proses ekstraksi karaginan dari rumput laut Kappaphycus alvarezii melalui variasi konsentrasi enzim selulase, konsentrasi kalsium hidroksida (Ca(OH)₂), dan suhu perendaman alkali dengan menggunakan metode Response Surface Methodology (RSM). Penelitian dilakukan menggunakan rancangan Box-Behnken RSM pada program Design Expert 13.0.5.0 dengan tiga variabel utama, yaitu konsentrasi enzim selulase (0; 0,55; dan 1%), konsentrasi Ca(OH)₂ (2; 5; dan 8%), serta suhu perendaman alkali (50; 60; dan 70°C). Hasil analisis menunjukkan bahwa formula optimum diperoleh pada nilai desirability sebesar 0,89 atau 89%, dengan komposisi optimum yaitu konsentrasi Ca(OH)₂ sebesar 2%, enzim selulase 0,6%, dan suhu perendaman alkali 70°C. Karakteristik karaginan yang dihasilkan meliputi rendemen 3,951%, kekuatan gel 931,978 g/cm², viskositas 198 cP, berat molekul 172,612 kDa, kadar air 11,936%, dan kadar abu 18,027%. | ||||||||||||
