Abstract

This study aims to examine the effect of explant type and cytokinin growth regulator type on shoot multiplication of Philodendron ‘Green Moss’ in vitro. This research was conducted at Esha Flora Plant Tissue Culture Laboratory, Bogor, West Java. A factorial Randomized Block Design (RBD) was used, with the two factor namely explant type and cytokinin type. The explant factor consisted of two levels, namely apical bud and lateral bud. The cytokinin factor consisted of five levels, namely control 0 mg/l; 2-iP 0.1 mg/l; TDZ 0.1 mg/l; BAP 0.1 mg/l and kinetin 0.1 mg/l. The results showed that both explant type and cytokinin type significantly influenced shoot multiplication. Apical bud explants significantly affected shoot length at 3-6 weeks after planting. Thidiazuron (TDZ) significantly affected plant height, number of leaves, number of buds, and bud length from 1-12 weeks after planting. No significant interaction was found between explant type and cytokinin type on the shoot formation of Philodendron ‘Green Moss’.

Additional Information: Penelitian ini bertujuan untuk mengkaji pengaruh jenis eksplan dan jenis zat pengatur tumbuh sitokinin terhadap multiplikasi tunas Philodendron ‘Green Moss’ secara in vitro. Penelitian ini dilaksanakan di Laboratorium Kultur Jaringan Tanaman Esha Flora, Bogor, Jawa Barat. Rancangan Acak Kelompok (RAK) faktorial digunakan dalam penelitian ini, dengan dua faktor yaitu jenis eksplan dan jenis sitokinin. Faktor eksplan terdiri atas dua taraf, yaitu tunas apikal dan tunas lateral. Faktor sitokinin terdiri atas lima taraf, yaitu kontrol 0 mg/l; 2-iP 0,1 mg/l; TDZ 0,1 mg/l; BAP 0,1 mg/l; dan kinetin 0,1 mg/l. Hasil penelitian menunjukkan bahwa baik jenis eksplan maupun jenis sitokinin berpengaruh nyata terhadap multiplikasi tunas. Eksplan tunas apikal memberikan pengaruh yang signifikan terhadap panjang tunas pada minggu ke-3 hingga minggu ke-6 setelah penanaman. Thidiazuron (TDZ) memberikan pengaruh signifikan terhadap tinggi tanaman, jumlah daun, jumlah tunas, dan panjang tunas dari minggu ke-1 hingga minggu ke-12 setelah penanaman. Tidak ditemukan interaksi yang signifikan antara jenis eksplan dan jenis sitokinin terhadap pembentukan tunas Philodendron ‘Green Moss’.
