Abstract
Among various types of seaweed, Caulerpa racemosa (Latoh) contains minerals and phytoconstituents that have potential as a bioreductor for silver nanoparticle (AgNPs) synthesis. This study aims to synthesize AgNPs using C. racemosa extract, examine the effect of AgNO₃ concentration (5 mM, 10 mM, 15 mM, and 20 mM), and determine the antibacterial capability of AgNPs products as active agents in chitosan film-forming solutions. The research process included four main stages: C. racemosa extraction, AgNPs synthesis, addition of AgNPs as active agents in chitosan film-forming solutions, and antibacterial testing of chitosan film-forming solutions. AgNPs were characterized using Fourier-transform Infrared Spectroscopy (FTIR), Scanning Electron Microscope (SEM), and antibacterial Well Diffusion analysis. Results showed that in FTIR analysis, C. racemosa extract contains -OH functional groups capable of reducing Ag⁺ ions to Ag⁰, making it effective as a bioreductor. Particle size characterized by SEM ranged from 217-224 nm, which was influenced by AgNO₃ concentration. Chitosan film-forming solutions showed high antibacterial activity against gram-negative bacteria, producing an average inhibition zone of 18 mm compared to 4 mm in pure chitosan films. C. racemosa shows great potential as an environmentally friendly natural bioreductor, and chitosan-AgNPs films as innovative materials that can be applied in the food, medical, and other industries.

| Additional Information: | Diantara berbagai jenis rumput laut, Caulerpa racemose (Latoh) memiliki kandungan mineral dan fitokonstituen yang potensial sebagai bioreduktor untuk sintesis nanopartikel perak (AgNPs). Penelitian ini bertujuan mensintesis AgNPs menggunakan ekstrak C. racemose, mengkaji pengaruh konsentrasi AgNO3 (5 mM, 10 mM, 15 mM, dan 20 mM), serta mengetahui kemampuan antibakteri pada produk AgNPs sebagai zat aktif larutan pembentuk film kitosan. Proses penelitian meliputi empat tahap utama: ekstraksi C. racemose, sintesis AgNPs, penambahan AgNPs sebagai zat aktif dalam larutan pembentuk film kitosan dan pengujian antibakteri larutan pembentuk film kitosan. AgNPs dikarakterisasi menggunakan Fourier-transform Infrared Spectroscopy (FTIR), Scanning Electron Microscope (SEM), dan antibacterial Well Diffusion analysis. Hasil menunjukkan bahwa pada analisa FTIR ekstrak C. racemose memiliki gugus fungsi -OH yang mampu mereduksi ion Ag+ menjadi Ag0, efektif sebagai bioreduktor. Ukuran partikel dikarakterisasi SEM berukuran range 217-224 nm yang dipengaruhi oleh konsentrasi AgNO3. Larutan pembentuk film kitosan menunjukkan aktivitas antibakteri pada bakteri gram-negatif yang tinggi, menghasilkan zona hambat rata-rata 18 mm dibandingkan 4 mm pada film kitosan murni. Potensi besar C. racemose sebagai bioreduktor alami yang ramah lingkungan dan film kitosan-AgNPs sebagai bahan inovatif yang dapat diaplikasikan dalam industri pangan, medis, dan lainnya. | ||||||
