Abstract

Anthracnose disease caused by Colletotrichum sp. fungus is one of the main obstacles in red chili cultivation because it causes serious damage to the fruit and reduces yields by more than 60%. The use of synthetic fungicides commonly has a negative impact on the environment and health, so that more environmentally friendly control alternatives are needed, through the use of rhizosphere bacteria as biological agents. This study aims to determine the potential of chili rhizosphere bacteria in inhibiting the growth of Colletotrichum sp. fungus in vitro, as well as to identify the morphological characteristics, gram properties, and pathogenicity of each isolate. This study used a non-factorial Completely Randomized Design, consisting of 7 treatments with 4 replications, resulting in a total of 28 experimental units. All isolates had antagonistic abilities against pathogenic fungi, with isolate BRC 6 showing the highest inhibition based on the diameter of the inhibition zone (8.37 mm) and the percentage of inhibition (7.81%). Morphological characterization and gram staining showed diversity between isolates, and in the potato pathogenicity test most of the bacterial isolates were not pathogenic. Thus, chili rhizosphere bacteria have the potential to be an effective and environmentally friendly biological agent in controlling anthracnose disease in vitro.

Additional Information: Penyakit antraknosa yang disebabkan oleh jamur Colletotrichum sp. merupakan salah satu kendala utama dalam budidaya cabai merah karena menyebabkan kerusakan serius pada buah dan menurunkan hasil panen hingga lebih dari 60%. Penggunaan fungisida sintetis yang umum dilakukan menimbulkan dampak negatif terhadap lingkungan dan kesehatan, sehingga diperlukan alternatif pengendalian yang lebih ramah lingkungan, melalui pemanfaatan bakteri rizosfer sebagai agen hayati. Penelitian ini bertujuan untuk mengetahui potensi bakteri rizosfer cabai dalam menghambat pertumbuhan jamur Colletotrichum sp. secara in vitro, serta mengidentifikasi karakteristik morfologi, sifat gram, dan patogenitas masing-masing isolat. Penelitian ini menggunakan Rancangan Acak Lengkap non-faktorial, yang terdiri dari 7 perlakuan dengan 4 ulangan, menghasilkan total 28 unit percobaan. Semua isolat memiliki kemampuan antagonis terhadap jamur patogen, dengan isolat BRC 6 menunjukkan daya hambat tertinggi berdasarkan diameter zona hambat (8,37 mm) dan persentase daya hambat (7,81%). Karakterisasi morfologi dan pewarnaan gram menunjukkan keragaman antar isolat, serta pada uji patogenitas kentang sebagian besar isolat bakteri tidak bersifat patogen. Sehingga bakteri rizosfer cabai memiliki potensi sebagai agensia hayati yang efektif dan ramah lingkungan dalam pengendalian penyakit antraknosa secara in vitro.
