Abstract

This research aimed to examine the effect of different concentrations of BAP (6-benzylaminopurine) and mung bean sprout extract on the in vitro growth of Philodendron Pink Princess shoots. The study was conducted from December 2024 to April 2025 at the Plant Physiology and Biotechnology Laboratory, Faculty of Agriculture, Sultan Ageng Tirtayasa University. A factorial Randomized Block Design was used, consisting of two factors: BAP concentrations (0, 1, 2, and 3 mg/L) and mung bean sprout extract concentrations (0, 25, 50, and 75 g/l). The parameters observed were shoot emergence time (weeks after planting/WAP), plant height (cm), number of leaves (leaves), number of shoots, percentage of browning, and percentage of contamination. The results showed that BAP had a highly significant effect on shoot emergence time and number of shoots. The fastest shoot emergence occurred with 1 mg/l BAP, while the highest number of shoots was observed at 2–3 mg/l BAP. Mung bean sprout extract had no significant effect on most parameters but significantly affected leaf number at 2 WAP and interacted with BAP in increasing leaf number at 12 WAP. The best combination for leaf number was b3t0 and b2t0. No browning symptoms were observed in any treatment (0%), and contamination occurred in 6.25% of explants, mainly in treatments with higher concentrations of sprout extract.

Additional Information: Penelitian ini bertujuan untuk mengkaji pengaruh berbagai konsentrasi BAP (6-benzilaminopurin) dan ekstrak tauge terhadap pertumbuhan tunas Philodendron Pink Princess secara in vitro. Penelitian dilaksanakan pada bulan Desember 2024 hingga April 2025 di Laboratorium Fisiologi dan Bioteknologi Tanaman, Fakultas Pertanian, Universitas Sultan Ageng Tirtayasa. Penelitian menggunakan Rancangan Acak Kelompok (RAK) faktorial yang terdiri dari dua faktor, yaitu konsentrasi BAP (0, 1, 2, dan 3 mg/L) dan konsentrasi ekstrak tauge (0, 25, 50, dan 75 g/L). Parameter yang diamati meliputi waktu muncul tunas (minggu setelah tanam/MST), tinggi tanaman (cm), jumlah daun (helai), jumlah tunas, persentase pencoklatan, dan persentase kontaminasi. Hasil penelitian menunjukkan bahwa BAP berpengaruh sangat nyata terhadap waktu muncul tunas dan jumlah tunas. Munculnya tunas tercepat terjadi pada perlakuan BAP 1 mg/L, sedangkan jumlah tunas terbanyak diperoleh pada perlakuan BAP 2–3 mg/L. Ekstrak tauge tidak berpengaruh nyata terhadap sebagian besar parameter, tetapi berpengaruh nyata terhadap jumlah daun pada 2 MST dan berinteraksi dengan BAP dalam meningkatkan jumlah daun pada 12 MST. Kombinasi terbaik untuk jumlah daun diperoleh pada perlakuan b3t0 dan b2t0. Tidak ditemukan gejala pencoklatan pada seluruh perlakuan (0%), dan kontaminasi terjadi sebesar 6,25% pada eksplan, terutama pada perlakuan dengan konsentrasi ekstrak tauge yang lebih tinggi.
