Abstract

The research was aimed to know shoot initiation of Kribo orchid (Dendrobium spectabile) with the addition of various concentrations of 6-benzylaminopurine (BAP) and sweet corn extract in vitro. The research was conducted at the Plant Physiology and Biotechnology Laboratory Agroecotechnology Department Faculty of Agriculture Sultan Ageng Tirtayasa University Serang Banten. A factorial Completely Randomized Design (CRD) was used in this research. The first factor was the BAP concentration, consisting of 4 levels: MS + BAP 0 ppm (B0), ½ MS + BAP 0 ppm (B1), ½ MS + BAP 1 ppm (B2) dan ½ MS + BAP 2 ppm (B3). The second factor was the concentration of sweet corn extract, consisting of 3 levels: 6% (E1), 9% (E2) dan 12% (E3), each replicated 3 times. The results showed that the concentration of ½ MS + BAP 1 ppm tended to have a better effect on the parameters of the number of shoots, explant height, and number of leaves. E1 6% concentration of sweet corn extract provided the best results for several parameters: number of shoots at 2 Week After Planting (WAP) (1 shoot), and 4 WAP (1.50 shoots); explant height at 2 WAP (1.60 cm), 4 WAP (2.02 cm), 6 WAP (2.28 cm), 8 WAP (2.38 cm) and 10 WAP (2.50 cm); and number of leaves at 2 WAP (7.58 leaves), 4 WAP (8.17 leaves), 6 WAP (8.75 leaves), 8 WAP (9.17 leaves), 10 WAP (9.75 leaves), and 12 WAP (10.42 leaves). There was no interaction between BAP concentration and sweet corn extract treatments on any of the observed parameters.

Additional Information: Penelitian ini bertujuan untuk mengetahui mengetahui tingkat keberhasilan inisiasi tunas anggrek kribo (Dendrobium spectabile) dengan penambahan tingkat konsentrasi 6 benzylaminopurine (BAP) dan ekstrak jagung manis secara in vitro. Penelitian dilaksanakan di di Laboratorium Fisiologi dan Bioteknologi Tanaman Agroekoteknologi Fakultas Pertanian Universitas Sultan Ageng Tirtayasa. Penelitian ini menggunakan Rancangan Acak Lengkap (RAL) faktorial yang terdiri dari dua faktor dan tiga ulangan. Faktor pertama adalah konsentrasi BAP (B), dengan 4 taraf, MS + BAP 0 ppm (B0), ½ MS + BAP 0 ppm (B1), ½ MS + BAP 1 ppm (B2) dan ½ MS + BAP 2 ppm (B3). Faktor kedua adalah konsentrasi ekstrak jagung manis (E) dengan 3 taraf, 6% (E1), 9% (E2) dan 12% (E3). Hasil penelitian menunjukkan bahwa perlakuan Konsentrasi BAP 1 ppm cenderung memberikan pengaruh lebih baik pada parameter jumlah tunas, tinggi eksplan dan jumlah daun. Konsentrasi 6% ekstrak jagung manis memberikan pengaruh terbaik pada paramter jumlah tunas 2 MST (1 tunas) dan 4 MST (1,50 tunas), pada parameter tinggi eksplan 2 MST (1,60 cm), 4 MST (2,02 cm), 6 MST (2,28 cm), 8 MST (2,38 cm), dan 10 MST (2,50 cm) dan pada parameter jumlah daun 2 MST (7,58 helai), 4 MST (8,17 helai), 6 MST (8,75 helai), 8 MST (9,17 helai), 10 MST (9,75 helai), dan 12 MST (10,42 Helai). Tidak terdapat interaksi antara perlakuan konsentrasi BAP dengan ekstrak jagung manis terhadap semua parameter yang diamati.
