Abstract
SUMMARY ISTI REKHA PERTIWI. Response of Using Culture Media and Different Concentration of Kinetin towards the Multiplication of Agarwood shoots (Aquilaria malaccensis) Using In Vitro way. (Guided by Susiyanti and Tb. Bahtiar Rusbana). The research was aimed to find out the response of using culture media and different concentration of kinetin towards the multiplication of Agarwood shoots using in vitro way. The research was conducted started from Mei until July 2013 in the Biotechnology Laboratorium of Faculty of Agriculture, Sultan Ageng Tirtayasa University, Serang, Banten. The research used Randomized Block Design that consisted of two factors. The first factor of culture media consisted of four levels: : M1 (Media MS); M2 (Media WPM); M3 (Media ½ MS); M4 (Media ½ WPM). The second factor, the concentration of kinetin, consisted of four levels: K0 (control); K1 (0,20 mg/l); K2 (0,40 mg/l); K3 (0,60 mg/l). Therefore, there were 16 combinations of treatments, each treatment was repeated three times so there were 48 experimental units. The result of the reseach has shown that the using of different culture media has significant effect in the parameter of plant height growth in 2 and 8 MST, the number of leaves in 2, 4, 6, 8 MST, the number of shoots in 4, 6, 8 MST and the number of sections in 2, 4, 6, 8 MST. The using of different concentrations of kinetin has given significant effects in the parameter of plant height growth in 2, 4, 6, 8 MST, the number of shoots in 8 MST and the number os sections in 4 and 8 MST. There were interaction between the culture media and the different concentrations of kinetin in parameter of plant height growth 2, 4, 6, 8 MST, the number of leaves in 2, 4, 4, 6, 8 MST, the number of shoots in 4, 6, 8, MST and the number of section in 4 MST. From the result of the research, it can be concluded that the treatment combination of MS media + 0,60 mg/ l kinetin is the best combination for the parameter of plant’s height growth, treatment combination MS media without addition of kinetin for the parameter of number of leaves, treatment combination media of ½ WPM + kinetin 0,40 mg/l fot the parameter of number of shoots and traetment combination media of ½ WPM without addition of kinetin for the parameter of number of sections.

Penelitian ini bertujuan untuk mengetahui respons penggunaan media kultur dan konsentrasi kinetin yang berbeda terhadap multiplikasi tanaman gaharu (Aquilaria malaccensis) secara in vitro. Penelitian ini dilaksanakan pada bulan Mei sampai bulan Juli 2013 di Laboratorium Bioteknologi Fakultas Pertanian Universitas Sultan Ageng Tirtayasa Serang, Banten. Percobaan ini menggunakan Rancangan Acak Kelompok (RAK) yang terdiri dari dua faktor. Faktor pertama media kultur, terdiri dari 4 taraf yaitu : M1 (Media MS); M2 (Media WPM); M3 (Media ½ MS); M4 (Media ½ WPM). Faktor kedua konsentrasi kinetin, terdiri dari 4 taraf yaitu : K0 (kontrol); K1 (0,20 mg/l); K2 (0,40 mg/l); K3 (0,60 mg/l). Sehingga terdapat 16 kombinasi perlakuan, setiap perlakuan diulang sebanyak 3 kali sehingga terdapat 48 satuan percobaan. Hasil penelitian menunjukkan bahwa perlakuan penggunaan media kultur yang berbeda berpengaruh nyata pada parameter pertambahan tinggi tanaman 2 dan 8 MST, jumlah daun pada 2, 4, 6, 8 MST, jumlah tunas pada 4, 6, 8 MST dan jumlah ruas pada 2, 4, 6, 8 MST. Perlakuan penggunaan konsentrasi kinetin yang berbeda memberikan pengaruh nyata pada parameter pertambahan tinggi tanaman pada 2, 4, 6, 8 MST, jumlah tunas pada 8 MST dan jumlah ruas pada 4 dan 8 MST. Terdapat interaksi antara media kultur dan konsentrasi kinetin yang berbeda pada parameter pertambahan tinggi tanaman 2, 4, 6, 8 MST, jumlah daun pada 2, 4, 4, 6, 8 MST jumlah tunas pada 4, 6, 8, MST dan jumlah ruas pada 4 MST. Dari hasil penelitian dapat disimpulkan bahwa kombinasi perlakuan media MS + kinetin 0,60 mg/l merupakan perlakuan terbaik untuk parameter pertambahan tinggi tanaman, kombinasi perlakuan media MS tanpa pertambahan kinetin untuk parameter jumlah daun, kombinasi perlakuan media ½ WPM + kinetin 0,40 mg/l untuk parameter jumlah tunas, dan kombinasi perlakuan media ½ WPM tanpa pertambahan kinetin untuk parameter jumlah ruas. | |||||||||
