Abstract
This research aim to determine the influence of MS (Murashige and Skoog) medium and BAP (Benzyl Amino Purine) Plant Growth Regulator (PGR) on the growthoforchid plants Anoectochilusformosanus. The researchwas conductedat the Plant Physiology and Biotechnology Laboratory, Faculty of Agriculture, Sultan Ageng Tirtayasa University. The planted orchids were observed for 12 Week After Planting (WAP). The method used was an experimental factorial Randomized Block Design (RBD). The trial consisted of eight combinations of treatments, each of which was repeated three times so that there were 24 units of trial. In one unit ofexperiment there are two bottles consisting ofone explanation so there are 48 explanations planted. The first factor was the concentration of MS (A), which consisted of 2 rate; A1 (½ MS Medium) and A2 (Full MS Medium). The second factor was that the PGR BAP (B) was comprised of 4 rate: B1 (0,25 ppm), B2 (0,50 ppm), B3 (1,00 ppm) and B4 (2,00 ppm). MS medium concentration treatment affected explant height parameters (6, 8, and 10 WAP). The A2 treatment (Full MS medium) gave the highest average result, which was 1,32 cm at the age of 12 WAP. The treatment of growth regulator concentrations of BAP had no realeffect onthe parameter number of shoots, explant height, and number of leaves from 0 to 12 WAP. There was an interaction between the treatment of MS medium concentration and BAP PGR at the age of 4 WAP with the highest average value in the A2B3 (Full MS Medium and 1 ppm BAP) which was 0,6 cm on the explant height parameter.

Additional Information: | Penelitian ini dilaksanakan pada bulan Januari sampai April 2024 di Laboratorium Fisiologi dan Bioteknologi Tanaman Fakultas Pertanian Universitas Sultan Ageng Tirtayasa Serang. Rancangan yang digunakan adalah Rancangan Acak Kelompok (RAK) faktorial. Faktor pertama adalah konsentasi media MS yang terdiri dari 2 taraf yaitu media ½ MS dan media MS full. Faktor kedua adalah konsentrasi zat pengatur tumbuh BAP yanag terdiri dari 4 taraf yaitu BAP 0,25 ppm; BAP 0,50 ppm; BAP 1 ppm; dan BAP 2 ppm. Parameter yang diamati yaitu persentase eksplan hidup (%), persentase eksplan browning, persentase eksplan terkontaminasi, jumlah tunas, tinggi eksplan (cm), dan jumlah daun (helai). Hasil penelitian menunjukkan bahwa perlakuan konsentrasi media MS berpengaruh terhadap parameter tinggi eksplan (6, 8, dan 10 MST). Perlakuan A2(media MS full) memberikan hasil rerata tertinggi yaitu 1,32 cm pada umur 12 MST. Perlakuan konsentrasi Zat Pengatur Tumbuh (ZPT) BAP tidak berpengaruh terhadap parameter jumlah tunas, tinggi eksplan dan jumlah daun dari 0 sampai 12 MST. Terdapat interaksi antara konsentrasi media MS dan ZPT BAP terhadap parameter tinggi eksplan pada umur 4 MST dengan nilai rerata tertinggi pada perlakuan A2B3 (media MS Full dan BAP 1 ppm) yaitu 0,6 cm. | |||||||||
