Abstract

Rhizosphere is a part of the soil located around the plant roots. The community of microorganisms in the rhizosphere is known to be more diverse than the nonrhizosphere soil. This research aimed to study the community of fungi in the rhizosphere of local rice varieties from Banten. Samples were collected from the rhizosphere of 8 local rice varieties of Banten was taken from 4 location in 2 districts, i.e. Lebak and Serang. Fungi were isolated using serial dilution method and plated in to Potato Dextrose Agar medium. The fungal isolates recovered from the medium were purified using the same medium, monosporic culture of the isolates then stored into cryo tube in 20% glycerol for long preservation. Fungal isolates were identified based on their morphological characters. In total, 35 isolates were recovered from 8 rhizosphere of local rice varieties of which belonged to 7 genera, namely Aspergillus, Chaetomium, Fusarium, Gongronella, Mucor, Penicillium, and Trichoderma. The most abundant rhizosphere fungal community was found in the varieties of Guling rice and Ruyung glutinous rice with 3 genera of 6 isolates obtained, those at the intermediate level was Limar rice with 2 genera out of 4 isolates, and the least found in Siang rice with 1 genus of 2 isolates. The result of this research was implemented on encyclopedia book as a learning source for high school students in grade X on the concept of fungi. The assessment of the encyclopedia book got a score of 85%. The assessment showed that the book is feasible to be used as a learning source. Keywords: Encyclopedia, Fungi, local rice, rhizosphere.

Additional Information: Rizosfer adalah bagian tanah yang terletak di sekitar akar tanaman. Komunitas mikroorganisme di rizosfer lebih beragam daripada di tanah non-rizosfer. Penelitian ini bertujuan untuk mempelajari komunitas fungi pada rizosfer varietas padi lokal Banten. Sampel dikumpulkan dari rizosfer 8 varietas padi lokal banten yang diambil dari 4 lokasi di 2 kabupaten, yaitu Lebak dan Serang. Fungi diisolasi menggunakan metode pengenceran serial dan dimasukkan ke dalam media Potato Dextrose Agar. Isolat fungi yang diperoleh dari media dimurnikan menggunakan media yang sama, kultur monosporik isolat kemudian disimpan ke dalam tabung cryo dalam gliserol 20% untuk penyimpanan yang lama. Isolat fungi diidentifikasi berdasarkan karakter morfologinya. Sebanyak 35 isolat diperoleh dari rizosfer 8 varietas padi lokal yang termasuk dalam 7 genera, yaitu Aspergillus, Chaetomium, Fusarium, Gongronella, Mucor, Penicillium, dan Trichoderma. Komunitas fungi rizosfer yang paling melimpah terdapat pada varietas padi Guling dan ketan Ruyung dengan didapatkan 3 genus dari 6 isolat, yang berada di tingkat menengah adalah padi Limar dengan didapatkan 2 genus dari 4 isolat, dan yang paling sedikit terdapat pada padi Siang dengan didapatkan 1 genus dari 2 isolat. Hasil penelitian ini diimplementasikan pada buku ensiklopedia sebagai sumber belajar siswa kelas X SMA tentang konsep fungi. Penilaian buku ensiklopedia mendapat skor 85%. Penilaian tersebut menunjukkan bahwa buku tersebut layak untuk digunakan sebagai sumber belajar. Kata kunci: Ensiklopedia, fungi, padi lokal, rizosfer.
