Abstract

Bananas, a major global food commodity, are threatened by Fusarium Tropical Race 4 (TR4), the causative agent of Panama disease. The study of the endophytic fungi diversity of Fusarium in wild bananas is crucial for exploring potential genetic resources. This research aims to identify and analyze the diversity of Fusarium endophytic fungi isolated from wild bananas (Musa sp.) from Sulawesi and Halmahera Islands. A total of 14 endophytic fungal isolates were obtained from 11 wild banana accessions and 1 cultivar accession using the single spore culture isolation technique. The identification process involved the observation of macroscopic and microscopic morphological characteristics, followed by molecular analysis, including DNA isolation, PCR with rpb2, tef-1α, and TR4 primers, and sequencing for species confirmation. Sequence analysis revealed a sequence similarity of over 90%. This research not only contributes to the preservation of bananas through the development of disease-resistant varieties, but also to the improvement of the quality of biology education through the implementation of research results based on the learning media of the monopoly game of the concept of biodiversity with the learning outcomes of students' critical thinking skills and understanding of science.

Additional Information: Pisang, komoditas pangan utama dunia, terancam oleh Fusarium Tropical Race 4 (TR4) penyebab Panama disease. Penelitian diversitas fungi endofit Fusarium pada pisang liar penting untuk eksplorasi sumber daya genetik potensial. Penelitian ini bertujuan untuk mengidentifikasi dan menganalisis diversitas fungi endofit genus Fusarium yang diisolasi dari pisang liar (Musa sp.) asal Pulau Sulawesi dan Halmahera. Sebanyak 14 isolat fungi endofit berhasil diperoleh dari 11 aksesi pisang liar dan 1 aksesi kultivar menggunakan teknik isolasi kultur spora tunggal. Proses identifikasi meliputi pengamatan karakteristik morfologi makroskopis dan mikroskopis, dilanjutkan dengan analisis molekuler melalui isolasi DNA, PCR menggunakan primer rpb2, tef-1α, dan TR4, serta sekuensing untuk konfirmasi spesies. Analisis sekuens menunjukkan tingkat kesamaan di atas 90%. Penelitian ini tidak hanya berkontribusi pada pelestarian pisang melalui pengembangan varietas tahan penyakit, tetapi juga pada peningkatan kualitas pendidikan biologi melalui implementasi hasil riset berbasis media pembelajaran permainan monopoli konsep biodiversitas dengan capaian pembelajaran keterampilan berpikir kritis dan pemahaman sains peserta didik.
