Abstract
Begonia is a plant with the potential to be an ornamental plant with high economic value. Biological control and increasing quality resistance in Begonia collection are needed for recovery and disease control using endophytic fungi. This research aims to identify endophytic fungi and their potential in controlling anthracnose pathogens by Colletotrichum sp. in the Begonia collection at the Green House of Begonia Bogor Botanical Gardens. The identification methods used morphological and molecular approaches for potential screening uses the in vitro dual culture method. Of the 24 strains, 9 genera of endophytic fungi were identified, including Absidia, Colletotrichum, Diaporthe, Humicola, Penicillium, Phyllosticta, Pyricularia, Mycelia sterilia, and Trichoderma. The highest inhibitory potential is shown by the Trichoderma (BGE 51 and BGE 72) with a percentage of 97%-100%, while the lowest potential is demonstrated by the Phyllosticta (BGE 20, BGE 24, and BGE 15) at 7%-36%. This research can enrich knowledge of the diversity of endophytic fungi in Begonia using specific ITS genes, thus providing more effective clues in controlling pathogens as biocontrol agents. The implications of this research are packaged as content recommendations presented in the form of learning videos that refer to the sub-concept of the fungal kingdom.

Additional Information: | Begonia merupakan salah satu tumbuhan yang berpotensi sebagai tanaman hias dengan nilai ekonomi tinggi. Pengendalian hayati dan peningkatan ketahanan mutu pada koleksi Begonia diperlukan untuk pemulihan dan pengendalian penyakit menggunakan fungi endofit. Penelitian ini bertujuan untuk mengidentifikasi fungi endofit dan potensinya dalam mengendalikan patogen antraknosa oleh Colletotrichum sp. pada koleksi Begonia di Green House of Begonia Kebun Raya Bogor. Metode identifikasi yang digunakan menggunakan pendekatan morfologi dan molekuler, kemudian untuk skrining potensi menggunakan metode dual culture secara in vitro. Dari 24 isolat telah teridentifikasi sebanyak 9 genus fungi endofit diantaranya Absidia, Colletotrichum, Diaporthe, Humicola, Penicillium, Phyllosticta, Pyricularia, Miselia sterilia, dan, Trichoderma. Potensi penghambatan tertinggi ditunjukan oleh genus Trichoderma (kode BGE 51 dan BGE 72) dengan persentase 97%-100%, sedangkan potensi terendah ditunjukan oleh genus Phyllosticta dan Pyricularia (kode BGE 20, BGE 24, dan BGE 15) sebesar 7%-36%. Penelitian ini dapat memperkaya pengetahuan akan diversitas fungi endofit pada Begonia menggunakan gen spesifik ITS sehingga memberikan petunjuk dalam mengendalikan patogen sebagai agen biokontrol dengan lebih efektif. Implikasi penelitian ini dikemas sebagai rekomendasi konten yang disajikan dalam bentuk video pembelajaran yang mengacu pada sub konsep kingdom fungi. | ||||||||||||
