Abstract

Oil palm (Elaeis guineensis) is the most demanded crop in the world because of the oil it produces. Due to the increasing number of enthusiasts every year, it is necessary to increase oil palm production. The main problem of oil palm is basal stem rot disease caused by Ganoderma boninense. Ganoderma prevention can be done in various ways, such as utilizing Ganoderma-resistant oil palm plants identified by Polymerase Chain Reaction (PCR) technique. This study aims to identify the cytosolic sulfotransferase 12-like (SOT12) resistance gene in two varieties of oil palm trees, MT-Gano (resistant) and Yangambi (susceptible), produced by PT Socfindo and to determine the closeness between the varieties used and the National Center for Biotechnology Information (NCBI) collection data. This research was conducted in the Agroindustry and Biomedical Laboratory of Badan Riset dan Inovasi Nasional (BRIN) Serpong from November 2023 to August 2024. This research used a qualitative descriptive method. The results showed that the SOT12 primer could be attached and reproduced but there were no significant differences between the two varieties. Analysis of blast results showed a similarity of 99.8% for resistant varieties and 99.9% for susceptible varieties with a coverage rate of 100% compared to several NCBI collection data. Phylogeny tree analysis proved that there is a dichotomy grouping of 60% in each variety, indicating that SOT12 can distinguish the pattern between the palm resistant and susceptible varieties to ganoderma.

Additional Information: Kelapa sawit (Elaeis guineensis) merupakan tanaman yang paling diminati di dunia karena minyak yang dihasilkannya. Oleh karena peminatnya meningkat setiap tahun maka perlu dilakukan peningkatan produktivitas kelapa sawit. Permasalahan utama kelapa sawit adalah penyakit busuk pangkal batang yang disebabkan oleh Ganoderma boninense. Pencegahan Ganoderma dapat dilakukan dengan berbagai cara, salah satunya dengan budidaya menggunakan bibit kelapa sawit unggul tahan Ganoderma yang dapat diketahui dengan teknik Polymerase Chain Reaction (PCR) dilanjutkan dengan sekuensing dengan metode Sanger. Penelitian ini bertujuan untuk mengidentifikasi gen ketahanan cytosolic sulfotransferase 12-like (SOT12) pada dua varietas kelapa sawit varietas MT-Gano (tahan) dan varietas Yangambi (rentan) hasil produksi PT. Socfindo dan mengetahui kedekatan antar verietas yang digunakan dibandingkan data koleksi National Center for Biotechnology Information (NCBI). Penelitian ini dilaksanakan di Laboratorium Agroindustri dan Biomedika, Badan Riset dan Inovasi Nasional (BRIN) Serpong dimulai bulan November 2023 sampai dengan Agustus 2024. Penelitian ini menggunakan metode deskriptif kualitatif. Hasil penelitian menunjukkan bahwa primer gen SOT12 dapat menempel dan diperbanyak melalui PCR namun tidak terlihat signifikan ukuran pita DNA antar varietas kelapa sawit tahan dan rentan. Hasil analisis Basic Local Alignment Search Tool (BLAST) pada data hasil sekuensing menunjukkan terdapat kesamaan sebesar 99,8% untuk varietas tahan dan 99,9% untuk varietas rentan dengan tingkat coverage 100% dengan beberapa data koleksi NCBI. Analisis pohon filogeni membuktikan terdapat pengelompokkan sebesar 60% pada setiap varietas yang menunjukkan bahwa SOT12 dapat membedakan pola kedua varietas.
