Abstract

The purpose of this study was to determine the effect of varying concentrations of growth regulators Benzyl Amino Purine (BAP) and Indole Acetic Acid (IAA) on the in vitro multiplication of Cattleya orchid shoots. The study was conducted at the Physiology and Biotechnology Laboratory, Department of Agroecotechnology, Faculty of Agriculture, Sultan Ageng Tirtayasa University, from April to July 2024. A Randomized Block Design was used, consisting of two factors. The first factor was the concentration of BAP at levels of 0 mg/l, 1 mg/l, and 2 mg/l, while the second factor was the concentration of IAA at levels of 0 mg/l, 0.1 mg/l, and 0.2 mg/l. The results showed that the absence of BAP significantly affected the number of roots at 12 weeks after planting (WAP), with an average of 1.41 roots. Similarly, the absence of IAA significantly affected the number of roots at 8 WAP, with an average of 0.83 roots. There was an interaction between the absence of both BAP and IAA (B0I0, control) on the explant height, resulting in the best height parameters at 8 WAP and 12 WAP, measuring 1.99 cm and 2.61 cm, respectively. The number of leaves for the B0I0 (control) treatment at 8 WAP was 10 leaves, while the B1I1 treatment (BAP 1 mg/l and IAA 0.1 mg/l) had 13 leaves at 12 WAP. The number of shoots for the B1I1treatment at 8 WAP was 3.5 shoots, and for the B0I0 (control) at 12 WAP, it was 4.25 shoots. The percentage of viable explants produced was 100%.

Additional Information: Tujuan penelitian ini adalah untuk mengetahui pengaruh variasi konsentrasi zat pengatur tumbuh Benzyl Amino Purine (BAP) dan Indole Acetic Acid (IAA) terhadap perbanyakan tunas anggrek Cattleya secara in vitro. Penelitian dilaksanakan di Laboratorium Fisiologi dan Bioteknologi, Jurusan Agroekoteknologi, Fakultas Pertanian, Universitas Sultan Ageng Tirtayasa, mulai bulan April sampai dengan Juli 2024. Penelitian yang digunakan adalah Rancangan Acak Kelompok yang terdiri atas dua faktor. Faktor pertama adalah konsentrasi BAP pada taraf 0 mg/l, 1 mg/l, dan 2 mg/l, sedangkan faktor kedua adalah konsentrasi IAA pada taraf 0 mg/l, 0,1 mg/l, dan 0,2 mg/l. Hasil penelitian menunjukkan bahwa tanpa pemberian BAP berpengaruh nyata terhadap jumlah akar pada umur 12 minggu setelah tanam (MST) dengan rata-rata 1,41 akar. Begitu pula tanpa pemberian IAA berpengaruh nyata terhadap jumlah akar pada umur 8 MST dengan rata-rata 0,83 akar. Terdapat interaksi antara tidak adanya BAP dan IAA (B0I0, kontrol) terhadap tinggi eksplan, sehingga menghasilkan parameter tinggi terbaik pada umur 8 MST dan 12 MST, yaitu masing-masing sebesar 1,99 cm dan 2,61 cm. Jumlah daun pada perlakuan B0I0 (kontrol) umur 8 MST sebanyak 10 helai, sedangkan pada perlakuan B1I1 (BAP 1 mg/l dan IAA 0,1 mg/l) berjumlah 13 helai pada umur 12 MST. Jumlah tunas pada perlakuan B1I1 umur 8 MST sebanyak 3,5 tunas, dan pada perlakuan B0I0 (kontrol) umur 12 MST sebanyak 4,25 tunas. Persentase eksplan viabel yang dihasilkan sebesar 100%.
