Abstract

Cellulose fiber is described as a string of fibers consisting of an amorphous phase and a crystalline phase, having a large surface area and high strength. Cellulose can be utilized as a material for regenerative medicine and as a biomaterial in the manufacture of hydrogel dressings. Cellulose can be extracted from Ulva lactuca seaweed. The success of cellulose extraction is highly dependent on the extraction method used. The cellulose fiber extraction used in this study is base hydrolysis with NaOH because it can remove lignin and increase cellulose content. The purpose of this study was to determine the optimum NaOH concentration and characteristics of cellulose fiber. This research was conducted in January - February 2023 at the Ecotoxicology Laboratory, National Research and Innovation Agency, Ancol. This study used a laboratory experimental method with a completely randomized design (CRD) consisting of one treatment factor, namely the concentration of NaOH solution (10%, 20%, 30%, 40%). This research consists of 4 stages, namely seaweed preparation, seaweed powder making, cellulose fiber extraction, and characterization. Parameters observed included Fourier Transform Infra Red (FTIR), X-ray diffraction (XRD), Simultaneous Thermal Analyzer (STA) and yield. FTIR, XRD, STA and yield test data were analyzed using quantitative descriptive method. The results showed that 30% NaOH concentration was the best treatment. Functional group analysis using FTIR showed absorption peaks of O-H O-H (3325,10 cm-1) and C-H (2912,3 cm-1). XRD results produced a diffraction peak at an angle of 2θ, which is 21,48°. The resulting yield was 4,38%. The type of U. lactuca seaweed cellulose was confirmed as α-cellulose and the degree of crystallinity was 84,093%.

Additional Information: Serat selulosa digambarkan sebagai untaian serat yang terdiri atas fase amorf dan fase kristal, memiliki permukaan yang luas dan kekuatan yang tinggi. Selulosa dapat dimanfaatkan sebagai bahan untuk pengobatan regeneratif dan sebagai biomaterial dalam pembuatan hydrogel dressing. Selulosa dapat diekstraksi dari rumput laut jenis Ulva lactuca. Keberhasilan ekstraksi selulosa sangat bergantung pada metode ekstraksi yang digunakan yaitu metode hidrolisis basa. Ekstraksi serat selulosa yang digunakan pada penelitian ini yaitu hidrolisis basa dengan NaOH karena dapat menghilangkan lignin dan peningkatan kandungan selulosa. Tujuan penelitian ini adalah menentukan konsentrasi NaOH yang optimum dan karakteristik serat selulosa. Penelitian ini dilaksanakan pada bulan Januari - Februari 2023 di Laboratorium Ekotoksikologi, Badan Riset dan Inovasi Nasional, Ancol. Penelitian ini menggunakan metode eksperimental laboratorium dengan Rancangan Acak Lengkap (RAL) terdiri dari satu faktor perlakuan yaitu perbedaan konsentrasi larutan NaOH (10%, 20%, 30%, 40%). Penelitian ini terdiri dari 4 tahapan yaitu preparasi rumput laut, pembuatan bubuk rumput laut, ekstraksi selulosa serat, dan karakterisasi. Parameter yang diamati meliputi, Fourier Transform Infra Red (FTIR), X-ray diffraction (XRD), Simultaneous Thermal Analyzer (STA) dan rendemen. Data uji FTIR, XRD, STA dan rendemen dianalisis menggunakan metode deskriptif kuantitatif. Hasil penelitian menunjukkan bahwa konsentrasi NaOH 30% adalah perlakuan terbaik. Analisis gugus fungsi menggunakan FTIR menunjukkan puncak serapan O-H (3325,10 cm-1) dan C-H (2912,3 cm-1). Hasil XRD menghasilkan puncak difraksi pada sudut 2θ, yaitu 21,48°. Rendemen yang dihasilkan sebesar 4,38%. Jenis selulosa rumput laut U. lactuca dikonfirmasikan sebagai α-selulosa dan derajat kristalinitas sebesar 84,093%.
