Abstract

Stevia rebaudiana a shrub originated from Paraguay, has been developed in many countries as a natural sweetener to substitute sugarcane. Previous researches focused on plantlet’s vigor by modifying macro nutrients contained in MS and WP media. MS and WP media used with macro nutrients at half strength, normal and double strength had unsatisfied results. The purpose of this research was to determine plantlet vigor characteristics of stevia BL and BP clones from meristem and ordinary cultures of apical and axillary shoots. The experimental design used was a completely randomized design (CRD) with 10 replications. The treatments consisted of 2 factors which were plantlets source: PM (apical shoot from meristem culture), SM (axillary shoot from meristem culture), PI (apical shoot from ordinary culture), SI (axillary shoot from ordinary culture), and stevia clones BL (BL clone), BP (BP clone). The results revealed that BP clone was more vigorous than BL clone because it had a significantly better response with plantlet height was 10,91 cm, the number of internude was 3,65, the number of shoots was 1,60, the number of roots was 7,07, and the highest in leaf size of 0,49 cm2. Axillary shoot from meristem culture clone BP was the best treatment with the highest plantlets of 12,57 cm at 4 WAP. Plantlets from axillary shoot of ordinary culture of clone BP had the highest in leaf size of 1,19 cm2. In general, the growth of plantlets derived from axillary shoots were better than that of apical shoots.

Additional Information: Stevia (Stevia rebaudiana Bertoni), termasuk tanaman perdu famili Astereceae berasal dari Paraguay. Seluruh bagian tanaman stevia kecuali akar mengandung glikosida yang memberikan rasa manis 200-300 kali dari gula tebu. Akumulasi glikosida tertinggi terdapat pada bagian daun. Teknik kultur jaringan dapat digunakan untuk menghasilkan bibit seragam secara massal dan cepat. Penelitian ini bertujuan untuk mengetahui vigor planlet stevia klon BL dan BP asal kultur meristem dan asal induk tanaman. Penelitian ini telah dilaksanakan pada bulan September sampai Desember 2016 di Laboratorium Biak Sel dan Mikropropagasi Tanaman, Pusat Penelitian Bioteknologi dan Bioindustri Indonesia, Bogor - Jawa Barat. Penelitian ini menggunakan Rancangan Acak Lengkap dengan 2 faktor, yaitu jenis planlet PM (Tunas pucuk asal kultur meristem), SM (Tunas samping asal kultur meristem), PI (Tunas pucuk asal induk tanaman), SI (Tunas samping asal induk tanaman), dan klon stevia BL (Klon BL), dan BP (Klon BP). Setiap perlakuan diulang sepuluh kali sehingga diperoleh 80 satuan percobaan. Parameter yang diamati adalah tinggi planlet, jumlah daun, jumlah ruas, jumlah tunas, jumlah akar, ukuran daun, diameter batang, tingkat kehijauan daun, bobot segar biomassa planlet, dan daya hidup planlet. Hasil penelitian menunjukkan bahwa klon BP lebih vigor dibanding klon BL karena secara signifikan memiliki respons yang lebih baik pada tinggi planlet 10,91 cm, jumlah ruas 3,65, jumlah tunas 1,60, jumlah akar 7,07, dan ukuran daun 0,49 cm2. Tunas samping asal kultur meristem klon BP merupakan perlakuan yang terbaik dalam hal tinggi planlet pada 4 MST dengan tinggi 12,57 cm. Tunas samping asal induk tanaman klon BP merupakan perlakuan yang terbaik dalam ukuran daun sebesar 1,19 cm2. Secara umum pertumbuhan planlet stevia asal tunas samping lebih baik dari asal tunas pucuk.
