Abstract
Banana is a plant that has many benefits and is in demand by the public, so it has quite prospective marketing prospects. However, the availability of quality banana seeds is limited and it takes a long time to get banana seeds, therefore a plant propagation method is used to produce seedlings in a relatively short time through the tissue culture method. The purpose of this study was to determine the level of growth regulator Thidiazuron (TDZ) concentrations in subcultures of two varieties of banana (Musa paradisiaca L.) in vitro. This research was conducted from April to July 2022 at the Physiology and Biotechnology Laboratory, Faculty of Agriculture, Sultan Ageng Tirtayasa University. In this study, a factorial randomized block design (RBD) was used. The first factor was banana varieties, consisting of 2 levels, namely Kepok Tanjung Bananas (P1) and Cavendish Grand Nain Bananas (P2). The second factor is the level of TDZ concentration which consists of 3 levels, namely 0,2 ppm (T1); 0,4 ppm (T2); 0,6 ppm (T3). The combination of the two treatments was repeated 3 times. The results showed that the Cavendish Grand Nain Banana Variety gave the best effect on the parameters of the percentage of living explants (100%), the percentage of contaminated explants (0%), and the number of roots (2,50 roots). TDZ concentration of 0,4 ppm tended to have a good effect on the time parameter of shoot emergence (1,24 MST). There was no interaction between the treatment of banana varieties and the level of TDZ concentrations in all observed parameters.

Pisang merupakan tanaman hortikultura yang memiliki banyak manfaat dan digemari oleh masyarakat Indonesia, sehingga memiliki prospek pemasaran yang cukup prospektif. Namun terbatasnya ketersediaan bibit pisang bermutu serta lamanya waktu yang dibutuhkan untuk mendapatkan bibit pisang, sehingga digunakan suatu metode perbanyakan tanaman untuk menghasilkan bibit dalam waktu yang relatif singkat melalui metode kultur jaringan. Penelitian ini bertujuan untuk mengetahui tingkat konsentrasi zat pengatur tumbuh Thidiazuron (TDZ) terhadap pertumbuhan tunas dua varietas pisang (Musa paradisiaca L.) secara in vitro. Penelitian ini dilaksanakan dari bulan April sampai bulan Juli 2022 di Laboratorium Fisiologi dan Bioteknologi, Fakultas Pertanian, Universitas Sultan Ageng Tirtayasa. Pada penelitian ini menggunakan Rancangan Acak Kelompok (RAK) faktorial. Faktor pertama yaitu varietas pisang, terdiri dari 2 taraf yaitu Pisang Kepok Tanjung (P1) dan Pisang Cavendish Grand Nain (P2). Faktor kedua yaitu tingkat konsentrasi TDZ yang terdiri dari 3 taraf yaitu 0,2 ppm (T1); 0,4 ppm (T2); 0,6 ppm (T3). Kombinasi kedua perlakuan tersebut diulang sebanyak 3 kali. Hasil penelitian menunjukkan bahwa varietas pisang cavendish grand nain memberikan pengaruh terbaik pada parameter persentase eksplan hidup (100%), persentase eksplan terkontaminasi (0%), dan jumlah akar (2,50 akar). Konsentrasi TDZ 0,4 ppm memberikan pengaruh cenderung baik pada parameter waktu muncul tunas (1,24 MST). Tidak terdapat interaksi antara perlakuan varietas pisang dengan tingkat konsentrasi TDZ pada semua parameter pengamatan. | |||||||||
