Abstract
The purpose on this study was to ascertain how the ideal concentration of TDZ (thidiazuron), a growth regulator, and MS media (Murashige & Skoog) affect the in vitro growth of Philodendron White Knight shoots. At the Biotechnology and Plant Physiology Laboratory, major in Agroecotechnology Faculty of Agriculture, Sultan Ageng Tirtayasa University, an experimental study was conducted from December 2023 to March 2024. A factorial Randomized Block Design (RBD) with two treatment components was employed in the study. The concentration level of Murashige and Skoog (MS) medium, which comes in two levels: ½ MS and MS was the first factor. The concentration level of thidiazuron (TDZ), which has four levels (T1 = 0.25 mg/l TDZ, T2 = 0.5 mg/l TDZ, T3 = 0.75 mg/l TDZ, and T41 = 1 mg/l TDZ. Shoot emergence time, plant height (cm), number of leaves (strands), percentage of sprouting explants (%), percentage of browning explants (%), and percentage of contamination (%) were the observation parameters recorded in this study. The results of the study indicated that the used of concentrations of 1 mg/l TDZ had a real influence on the time the shoots appeared and had a very influence on the plant height, used of concentrations of 0.25 mg/l TDZ had a very influence number of leaves as well as the addition of Plant Preservative Mixture (PPM) was successful in preventing contamination (0%) and browning (0%) on the growts of explants.

Additional Information: | Philodendron termasuk keluarga Araceae, yang pertumbuhannya lambat dan jarang sekali menghasilkan bunga. Perbanyakan tanaman ini biasanya dilakukan secara vegetatif dengan cara memotong batang utamanya. Perbanyakan secara in vitro dapat menghasilkan bibit Philodendron unggul dalam jumlah banyak, seragaam dan tidak terkendala iklim, dengan cara menambahkan ke dalam media yaitu zat pengatur tumbuh Thidiazuron (TDZ) dalam bah konsentrasi rendah (0,1- 1,5 mg/l). Penelitian bertujuan untuk mengetahui pengaruh konsentrasi terbaik media MS (Murashige & Skoog) dan zat pengatur tumbuh TDZ (thidiazuron) terhadap pertumbuhan tunas philodendron White Knight secara in vitro. Penelitian ini merupakan penelitian eksperimen yang dilaksanakan pada bulan Desember 2023 sampai Maret 2024 yang bertempat di Laboratorium Bioteknologi dan Fisiologi Tanaman, Jurusan Agroekoteknologi Fakultas Pertanian Universitas Sultan Ageng Tirtayasa. Penelitian menggunakan Rancangan Acak Kelompok (RAK) faktorial terdiri dari 2 faktor perlakuan. Faktor pertama adalah tingkat konsentrasi media Murashige & Skoog (MS) yang terdiri dari dua taraf, yaitu : ½ MS dan MS. Faktor kedua adalah tingkat konsentrasi thidiazuron (TDZ) yang terdiri dari empat taraf, yaitu : T1 = 0,25 mg/l TDZ, T2 = 0,5 mg/l TDZ, T3 = 0,75 mg/l TDZ, T4 = 1 mg/l TDZ. Parameter pengamatan dalam penelitian adalah waktu muncul tunas, tinggi tanaman (cm), jumlah daun (helai), persentase eksplan bertunas (%), persentase eksplan browning (%), dan persentase kontaminasi (%). Hasil penelitian menunjukkan bahwa konsentrasi 1 mg/l TDZ berpengaruh nyata terhadap waktu muncul tunas dan berpengaruh sangat nyata terhadap tinggi tanaman, konsentrasi 0,25 mg/l TDZ berpengaruh sangat nyata terhadap jumlah daun serta penambahan Plant Preservative Mixture (PPM) berhasil mencegah kontaminasi (0%) dan browning (0%) pada pertumbuhan eksplan. | |||||||||
