Abstract
This study aims to determine the effect of antibiotic type and soaking time on the percentage of sterile explants on Jaboticaba (Plinia cauliflora) plants in vitro. This study used a factorial randomized design which consisted of two factors, namely the type of antibiotic and soaking duration of the explants. The type of antibiotic factor consists of 4 levels, namely Propolis, Amoxicillin, Streptomycin, and Amoxicillin + Streptomycin. Meanwhile, the soaking time factor consists of 3 levels, namely soaking for 1 hour, soaking for 12 hours, and soaking for 24 hours. The results of the study showed that the type of antibiotic treatment and the length of soaking gave results that were not significantly different on the precentage of sterille explants. This is different from the interaction treatment which has a very significant different effect on the percentage of sterile explants of Jaboticaba leaf. Soaking the explants in the Amoxicillin antibiotic solution gave the best percentage of sterile explants namely 83.33%, while for the explant soaking time treatment, the largest percentage of sterile explants, namely 29,17% was found in the 24 hour explant soaking treatment.

| Additional Information: | Tanaman Jaboticaba (Plinia cauliflora) merupakan tanaman buah endemik yang berasal dari hutan di Brazil. Tanaman ini memiliki potensi tinggi jika dikembangkan, tetapi akibat lamanya pertumbuhan tanaman ini maka dapat dipersingkat melalui perbanyakan secara in vitro. Untuk mengurangi tingkat browning yang disebabkan oleh tingginya kadar senyawa fenolik pada tanaman maka digunakan media WPM (Wood Plant Media). Penelitian ini bertujuan untuk mengetahui pengaruh jenis antibiotik dan lama perendaman terhadap persentase eksplan steril pada tanaman Jaboticaba (Plinia cauliflora) secara in vitro. Penelitian ini menggunakan Rancangan Acak Kelompok (RAK) Faktorial yang terdiri atas dua faktor, yaitu jenis antibiotik dan lama perendaman eksplan. Faktor jenis antibiotik terdiri dari 4 taraf, yaitu Propolis, Amoxicillin, Streptomycin, dan Amoxicillin + Streptomycin. Sedangkan faktor lama perendaman terdiri dari 3 taraf, yaitu perendaman selama 1 jam, perendaman selama 12 jam, dan perendaman selama 24 jam. Hasil penelitian menunjukkan bahwa perlakuan jenis antibiotik serta lama perendaman memberikan hasil yang tidak berbeda nyata. Berbeda dengan perlakuan interaksi yang memiliki pengaruh berbeda sangat nyata terhadap persentase eksplan steril daun Jaboticaba. Perendaman eksplan kedalam larutan antibiotik Amoxicillin memberikan persentase eksplan steril terbaik yaitu sebesar 83,33%, untuk perlakuan lama perendaman eksplan persentase eksplan steril terbesar yaitu 29,17% terdapat pada perlakuan perendaman eksplan selama 24 jam. | ||||||||||||
