Abstract

Pulmonary Tuberculosis (TB) is an infectious disease that affects the lungs and is caused by the bacteria Mycobacterium tuberculosis where according to data from the World Health Organization (WHO), the number of TB patients continues to increase from year to year. The treatment of tuberculosis has undergone developments related to the use of more effective drugs where currently a combination of rifampicin along with other drugs is used which can treat pulmonary tuberculosis cavities after 3 to 6 months of use. Sample extraction was carried out by maceration method using methanol solvent for 3 x 24 hours. Furthermore, the crude methanol extract was carried out qualitative and quantitative tests of flavonoid content and analyzed by toxicity tests with BSLT testing, GC-MS testing and molecular tethering in silico. The results of this study are the yield of the extract of 2.37%, has a positive response to the presence of flavonoids marked by the formation of orange color and has a content of 642.70 ppm, identified as many as 33 compounds contained, has a toxicity level with an LC50 value of 36.38 ppm, has potential as a TB protein inhibitor with the compound 1,7,7-Trimethyl-3-phenethylidenebicyclo[2. 2.1]heptan-2-one with binding affinity value on protein 4FDO of -7 kcal/mol, 4FEH of -6.8 kcal/mol and on 4NCR of -8.6 kcal/mol.

Additional Information: Tuberkulosis Paru (TBC) adalah penyakit infeksi yang menyerang paru-paru dan disebabkan oleh bakteri Mycobacterium tuberculosis di mana menurut data Badan Kesehatan Dunia (WHO), jumlah penderita TBC terus meningkat dari tahun ke tahun. Pengobatan TBC telah mengalami perkembangan terkait penggunaan obat yang lebih efektif di mana pada saat ini digunakan kombinasi rifampisin bersama dengan obat lain yang dapat mengobati kavitas tuberkulosis paru setelah 3 sampai 6 bulan penggunaannya. Ekstraksi sampel dilakukan dengan metode maserasi dengan menggunakan pelarut metanol selama 3 x 24 jam. Selanjutnya crude ekstrak methanol dilakukan uji kualitatif dan kuantitatif kandungan flavonoid dan dianalisa dengan uji toksisitas dengan BSLT testing, pengujian GC-MS dan penambatan molekul secara in silico. Hasil dari penelitian ini adalah didapatkan rendemen dari ektrak sebesar 2,37%, memiliki respon positif terhadap keberadaan flavonoid yang ditandai dengan terbentuknya warna jingga dan memiliki kandungan sebesar 642,70 ppm, teridentifikasi sebanyak 33 senyawa yang terkandung, memiliki tingkat toksisitas dengan nilai LC50 sebesar 36,38 ppm, memiliki potensi sebagai inhibitor protein TBC dengan senyawa 1,7,7-Trimethyl-3-phenethylidenebicyclo[2.2.1]heptan-2-one dengan nilai binding affinity pada protein 4FDO sebesar -7 kcal/mol, 4FEH sebesar -6,8 kcal/mol dan pada 4NCR sebesar -8,6 kcal/mol.
