Abstract
This study aims to determine the effectiveness of exposure time and concentration of 2,4-D (Dichlorophenoxyacetic Acid) against Si Seupah durian callus induction (Durio zibethinus Murr.) respectively in vitro. This research was carried out at the Plant Physiology and Biotechnology Laboratory, Department of Agroecotechnology, Faculty of Agriculture, Sultan Ageng Tirtayasa University. The research design used the Split-Plot design, which consists of 2 factors. The first factor or main plot was the photoperiod with 2 levels (16 and 24 hours). The second factor or subplot was the 2,4-D concentration, which consists of 5 levels (0; 1; 2; 3; and 4 ppm). The results showed that the 2,4-D concentration treatment had a significant on the time the callus appeared, and had the effect of increasing the callus area by (0.0003 mm2). There was no interaction between the photoperiod and the 2,4-D. In this study, the explants that were able to form callus had a compact texture, the color of the callus was predominantly pale yellow, the percentage of explants browning was 50%, contaminated explants were 0%, and the percentage of live explants was 73.33%.

Additional Information: | Penelitian ini bertujuan untuk mengetahui efektivitas lama penyinaran dan konsentrasi 2,4-D (Dichlorophenoxyacetic Acid) terhadap induksi kalus durian Si Seupah (Durio zibethinus Murr.) secara in vitro. Penelitian ini dilaksanakan di Laboratorium Fisiologi dan Bioteknologi Tanaman, Jurusan Agroekoteknologi, Fakultas Pertanian, Universitas Sultan Ageng Tirtayasa. Rancangan penelitian yang digunakan yaitu Rancangan Petak Terbagi yang terdiri dari 2 faktor. Faktor pertama atau petak utama adalah lama penyinaran dengan 2 taraf yaitu 16 dan 24 jam. Faktor kedua atau anak petak adalah konsentrasi 2,4-D yang terdiri dari 5 taraf, yaitu 0; 1; 2; 3; dan 4 ppm. Hasil penelitian menunjukkan bahwa perlakuan konsentrasi 2,4-D memberikan pengaruh nyata terhadap waktu muncul kalus, dan memberikan pengaruh penambahan luasan kalus sebesar (0,0003 mm2). Tidak ada interaksi antara perlakuan lama penyinaran dengan konsentrasi 2,4-D. Pada penelitian ini eksplan yang mampu membentuk kalus memiliki tekstur yang kompak, warna kalus didominasi kekuningan, persentase eksplan browning sebesar 50%, eksplan yang terkontaminasi 0%, dan persentase eksplan hidup sebesar 73,33%. | |||||||||
