Abstract

Passion fruit plants are important economically due to the high nutrient value of the fruit, and they have an important role in the pharmaceutical world. People often exploit passion fruit plants for medicinal purpose without cultivating them. Passion fruit propagation can be done using seeds, grafting, of cuttings. However, such propagation can increase the value of high genetic variability so that to overcome this can be done by propagating passion fruit seedlings in tissue cultue by callus induction. This research was carried out to determine the effect of 2,4-D and BAP on callus induction in Ciangir passion fruit leaves. The method utilized was a factorial Randomized Group Design (RGD), which consisted of two factors, namely the 2,4-D concentration level (0, 1, 2, 3) ppm and the BAP concentration level (0, 1, 2) ppm, so there were 12 treatment combinations which were repeated three times. The observed parameters were the percentage of callus growth time, callus area, percentage of callus explants, percentage of contaminated explants, percentage of live explants, percentage of browning explants, callus color and texture, percentage of globular phase, percentage of heart phase, percentage of torpedo phase, and percentage of cotyledon phase. The concentration level of 2,4-D 3 ppm and BAP 0 ppm best affected the callus area parameter of 15.179,33 mm^2 . The results showed that the percentage of the fastest callus emergence time at 2 week after culture was 22,2%, the percentage of the highest callus explant was 100%, the percentage of the highest contaminated explants was 0%, the percentage of the highest living explants was 100%, the percentage of the highest browning explants was 66%, the color of callus was green to yellowish, the texture of callus was crumbly, compact, and intermediate, the percentage of the highest globular phase was 33,33%, the percentage of the highest heart phase was 33,33%, the percentage of the highest torpedo phase was 66,67%% and the percentage of the highest cotyledone phase was 0%. The research should focus on callus induction of ciangir passion fruit leaves with other growth regulators.

Additional Information: Tanaman markisa penting secara ekonomi karena nilai gizi buahnya yang tinggi, dan mempunyai peranan penting dalam dunia farmasi. Masyarakat sering memanfaatkan tanaman markisa untuk tujuan pengobatan tanpa membudidayakannya. Perbanyakan buah markisa dapat dilakukan dengan menggunakan biji, okulasi, atau stek. Namun perbanyakan tersebut dapat meningkatkan nilai variabilitas genetik yang tinggi sehingga untuk mengatasinya dapat dilakukan dengan perbanyakan bibit markisa pada kultur jaringan dengan induksi kalus. Penelitian ini dilakukan untuk mengetahui pengaruh 2,4-D dan BAP terhadap induksi kalus daun markisa Ciangir. Metode yang digunakan adalah Rancangan Acak Kelompok (RAK) faktorial yang terdiri dari dua faktor yaitu taraf konsentrasi 2,4-D (0, 1, 2, 3) ppm dan taraf konsentrasi BAP (0, 1, 2). ppm sehingga terdapat 12 kombinasi perlakuan yang diulang sebanyak tiga kali. Parameter yang diamati adalah persentase waktu tumbuh kalus, luas kalus, persentase eksplan kalus, persentase eksplan terkontaminasi, persentase eksplan hidup, persentase eksplan pencoklatan, warna dan tekstur kalus, persentase fase globular, persentase fase jantung, persentase fase torpedo, dan persentase fase kotiledon. Tingkat konsentrasi 2,4-D 3 ppm dan BAP 0 ppm paling baik mempengaruhi parameter luas kalus sebesar 15.179,33 mm^2. Hasil penelitian menunjukkan persentase waktu munculnya kalus tercepat pada minggu ke 2 setelah budidaya sebesar 22,2%, persentase eksplan kalus tertinggi sebesar 100%, persentase eksplan terkontaminasi tertinggi sebesar 0%, persentase eksplan terkontaminasi tertinggi sebesar 0%. eksplan hidup sebesar 100%, persentase eksplan berwarna coklat tertinggi sebesar 66%, warna kalus hijau sampai kekuningan, tekstur kalus gembur, kompak dan sedang, persentase fase globular tertinggi sebesar 33,33% , persentase fase jantung tertinggi sebesar 33,33%, persentase fase torpedo tertinggi sebesar 66,67%% dan persentase fase kotiledon tertinggi sebesar 0%. Penelitian sebaiknya fokus pada induksi kalus daun markisa ciangir dengan zat pengatur tumbuh lainnya.
