Abstract
This research is aimed to determine the effect of various types of media and auxin growth regulators on the callus induction of kokoleceran (Vatica bantamensis) leaf in vitro. The research was carried out from June to November 2023 at the Plant Physiology and Biotechnology Laboratory, Faculty of Agriculture, Sultan Ageng Tirtayasa University. The method used was an experimental factorial Randomized Complete Block Design (RCBD). The first factor was types of media consisting of 2 levels, namely MS media and WPM media. The second factor was the variation of auxin growth regulators which consists of 6 levels, namely 1 ppm Picloram, 3 ppm Picloram, 5 ppm Picloram, 1 ppm Picloram + 1 ppm 2,4-D, 3 ppm Picloram + 1 ppm 2,4-D and 5 ppm Picloram + 1 ppm 2,4-D. The result showed that both treatments had no significant influence on all of the parameters observed. Callus diameter and callus area parameter at the age of 6 WAP (Week After Planting) with WPM media produces an average value that tends to show better results (namely at 1,05 mm and 1,13 mm2). The treatment of auxin Plant Growth Regulator (PGR) 5 ppm Picloram + 1 ppm 2,4-D produced an average value that tends to show better result for the callus diameter and callus area parameter at 6 WAP (namely 1,07 mm and 1,18 mm2).

Additional Information: | Penelitian ini bertujuan untuk mengetahui pengaruh variasi jenis media dengan zat pengatur tumbuh auksin terhadap induksi kalus daun kokoleceran (Vatica bantamensis) secara in vitro. Penelitian ini dilaksanakan pada bulan Juni sampai November 2023 di Laboratorium Fisiologi dan Bioteknologi Tanaman Fakultas Pertanian Universitas Sultan Ageng Tirtayasa Serang. Rancangan yang digunakan adalah Rancangan Acak Kelompok (RAK) faktorial. Faktor pertama adalah jenis media yang terdiri dari 2 taraf yaitu media MS dan media WPM. Faktor kedua adalah zat pengatur tumbuh (ZPT) auksin yang terdiri dari 6 taraf yaitu 1 ppm Pikloram, 3 ppm Pikloram, 5 ppm Pikloram, 1 ppm Pikloram + 1 ppm 2,4-D, 3 ppm Pikloram + 1 ppm 2,4-D dan 5 ppm Pikloram + 1 ppm 2,4-D. Parameter yang diamati yaitu waktu muncul kalus, diameter kalus, luas kalus, warna kalus, tekstur kalus, persentase eksplan berkalus, persentase eksplan browning, dan persentase kontaminasi. Hasil penelitian menunjukkan bahwa kedua perlakuan tidak memberikan pengaruh nyata terhadap seluruh parameter yang diamati. Parameter diameter kalus dan luas kalus pada umur 6 MST dengan media WPM menghasilkan nilai rata-rata yang cenderung lebih besar yaitu sebesar 1,05 mm dan 1,13 mm2. Sedangkan pada perlakuan ZPT auksin 5 ppm Pikloram + 1 ppm 2,4-D menghasilkan nilai rata-rata yang cenderung lebih besar untuk parameter diameter kalus dan luas kalus pada 6 MST yaitu sebesar 1,07 mm dan 1,18 mm2. | |||||||||
