Abstract

Glucomannan is a water-soluble polysaccharide contained in porang tubers. Ethanol extraction can be used to extract glucomannan from fresh porang tubers. This research aimed to perform physicochemical characterization of glucomannan flour extracted from fresh porang tubers. Randomized Complete Block Design (RCBD) is used with one factor, namely various extraction treatments. A total of ten treatments that were repeated twice analyzed for its yield, glucomannan content, oxalate content, color, whiteness index, viscosity, water content, and ash content. The result showed that differentiating extraction treatments have significant effect on glucomannan content, oxalate content, b* chromaticity, whiteness index, water content, and ash content. Based on physicochemical characteristics analyzed, two treatments were selected using Bayes method, they were three-stage ethanol extraction using graded concentrations of 50, 70, then 96% with addition of one-hour stirrer treatment and one-stage ethanol extraction using a concentration of 70% with addition of two-hours stirrer treatment. These treatments then analyzed further for protein content, fat content, carbohydrate content, functional group characterization using FTIR, and surface morphology using SEM. Glucomannan contained in the first selected treatment is 93,412% while the second selected treatment is 92,697%. Each treatment has the characteristics of water content 5,366 and 5,985%, ash content 0,852 and 1,413%, fat content 0,245 and 0,525%, protein content 1,735 and 2,845%, carbohydrate content 91,802 and 89,872%, oxalate content 185,185 and 44,444%, L* chromaticity 79,60 and 79,05, a* chromaticity 1,52 and 1,65, b* chromaticity 16,05 and 28,40, whiteness index 73,983 and 64,670, yield 12,743 and 12,386%, and viscosity 997,0 and 969,8 mPa·s.

Additional Information: Glukomanan merupakan polisakarida larut air yang terkandung dalam umbi porang. Ekstraksi etanol dapat digunakan untuk mengekstraksi glukomanan dari umbi porang segar. Penelitian ini bertujuan untuk mengkarakterisasi fisikokimia tepung glukomanan yang diekstraksi dari umbi porang segar. Penelitian menggunakan Rancangan Acak Kelompok Lengkap (RAK) satu faktor yaitu perbedaan perlakuan ekstraksi. Sepuluh perlakuan diulang sebanyak dua kali, lalu dianalisis rendemen, kadar glukomanan, kadar oksalat, warna dan derajat putih, viskositas, kadar air, dan kadar abu. Hasil penelitian menunjukkan bahwa perbedaan perlakuan ekstraksi memberikan pengaruh nyata terhadap kadar glukomanan, kadar oksalat, kromatisitas b*, indeks keputihan, kadar air, dan kadar abu. Berdasarkan sifat fisikokimia yang dianalisis, dipilih dua perlakuan dengan menggunakan metode Bayes, yaitu ekstraksi etanol tiga tingkat dengan konsentrasi bertingkat 50, 70, lalu 96% dengan penambahan perlakuan pengadukan selama satu jam dan ekstraksi etanol satu tingkat dengan konsentrasi 70% dengan penambahan perlakuan pengadukan selama dua jam. Perlakuan tersebut kemudian dianalisis lebih lanjut kadar protein, kadar lemak, kadar karbohidrat, karakterisasi gugus fungsi menggunakan FTIR, dan morfologi permukaannya menggunakan SEM. Kandungan glukomanan pada perlakuan terpilih pertama sebesar 93,412% sedangkan pada perlakuan terpilih kedua sebesar 92,697%. Masing-masing perlakuan mempunyai karakteristik kadar air 5,366 dan 5,985%, kadar abu 0,852 dan 1,413%, kadar lemak 0,245 dan 0,525%, kadar protein 1,735 dan 2,845%, kadar karbohidrat 91,802 dan 89,872%, kadar oksalat 185,185 dan 44,444%, L* kromatisitas 79,60 dan 79,05, kromatisitas a* 1,52 dan 1,65, kromatisitas b* 16,05 dan 28,40, derajat putih 73,983 dan 64,670, rendemen 12,743 dan 12,386%, serta viskositas 997,0 dan 969 ,8 mPa·s.
