Abstract
Mangosteen Macakal is one of the superior varieties originating from Banten Province. This research was aimed to determine the effect of 2,4-D and BAP concentration on the initiation of embryogenic callus of mangosteen (Garcinia mangostana L.) var. Macakal by in vitro. This research was conducted at the Laboratory Biotechnology, Sultan Ageng Tirtayasa University, Serang, Banten Province from June-Oktober 2023. The research used a randomized block design, consisted two factors and four replications. The first factor was the concentration of 2,4-D (P) which consisted of two levels, namely: 1 ppm (P1) and 2 ppm (P2). The second factor was the concentration of BAP (R) which consisted of three levels, namely: 0,5 ppm (R1), 0,75 ppm (R2), and 1 ppm (R3). The result showed that the treatment of 2,4-D there is no significant effect on callus diameter, callus area and embryogenic callus growing. The treatment of BAP gave the best results on the callus diameter (0,79 cm). then on the callus area (0,97 cm2) in the 12 WAP.

Additional Information: | Manggis Macakal merupakan salah satu varietas unggul yang berasal dari Provinsi Banten. Penelitian ini bertujuan untuk mengetahui pengaruh konsentrasi 2,4-D dan BAP terhadap inisiasi kalus embriogenik tanaman manggis (Garcinia mangostana L.) var. Macakal secara in vitro. Penelitian ini dilaksanakan di Laboratorium Bioteknologi Universitas Sultan Ageng Tirtayasa, Serang, Provinsi Banten pada bulan Juni-Oktober 2023. Penelitian menggunakan rancangan acak kelompok, terdiri dari dua faktor dan empat ulangan. Faktor pertama adalah konsentrasi 2,4-D (P) yang terdiri dari dua taraf yaitu: 1 ppm (P1) dan 2 ppm (P2). Faktor kedua adalah konsentrasi BAP (R) yang terdiri dari tiga taraf yaitu: 0,5 ppm (R1), 0,75 ppm (R2), dan 1 ppm (R3). Hasil penelitian menunjukkan bahwa perlakuan 2,4-D tidak memberikan pengaruh nyata terhadap diameter kalus, luasan kalus dan pertumbuhan kalus embriogenik. Perlakuan BAP memberikan hasil terbaik pada diameter kalus (0,79 cm). kemudian pada luasan kalus (0,97 cm2) pada umur 12 MST. | |||||||||
