Abstract

Miracle Fruit is a type of tropical plant native to West Africa which is included in the Sapotaceae family. Miracle fruit is a unique fruit that can change the taste of food become sweet. Miracle fruit has many benefits, as a natural food sweetener, diabetes medicine, and gardening plant. This study aims to determine the effect of clorox concentration and the length of soaking clorox on the initiation of miracle fruit seed (Synsepalum dulcificum) in vitro. This research was conducted from January to April 2023 at the Esha Flora Plant and Tissue Culture Laboratory, Bogor, West Java. This research used a Completely Randomized Design (CRD) with 2 factors. The first factor is the clorox concentration with 4 levels (0%, 15%, 20%, and 25%) and the second factor is the length of soaking clorox soaking with 4 levels (5, 10, 15, and 20 minutes). The results of the study showed that the clorox concentration level treatment had a significant effect on the time the seed explants were contaminated with bacteria and had a very significant effect on the percentage of seed explants contaminated with bacteria and germinating, as well as the time the seed explants germinated. In this study, the best seed explant treatment is at 0% clorox concentration because shows more seed explants germinated. The length of soaking in clorox does not have a significant effect on the seed initiation of miracle fruit, but it can reduce the percentage of contamination by 83.33% and the percentage of browning by 100%.

Additional Information: Miracle Fruit merupakan salah satu jenis tanaman tropis asli Afrika Barat yang termasuk dalam famili Sapotaceae. Miracle fruit merupakan buah unik yang mampu mengubah rasa makanan menjadi manis. Miracle fruit mempunyai banyak manfaat, sebagai pemanis makanan alami, obat diabetes, dan tanaman pekarangan. Penelitian ini bertujuan untuk mengetahui pengaruh konsentrasi clorox dan lama perendaman clorox terhadap inisiasi biji miracle fruit (Synsepalum dulcificum) secara in vitro. Penelitian ini dilaksanakan pada bulan Januari hingga April 2023 di Laboratorium Tanaman dan Kultur Jaringan Esha Flora, Bogor, Jawa Barat. Penelitian ini menggunakan Rancangan Acak Lengkap (RAL) dengan 2 faktor. Faktor pertama adalah konsentrasi clorox dengan 4 level (0%, 15%, 20%, dan 25%) dan faktor kedua adalah lama perendaman clorox dengan 4 level (5, 10, 15, dan 20 menit). Hasil penelitian menunjukkan bahwa perlakuan tingkat konsentrasi clorox memberikan pengaruh nyata terhadap waktu eksplan biji terkontaminasi bakteri dan memberikan pengaruh sangat nyata terhadap persentae eksplan biji terkontaminasi bakteri dan berkecambah, serta waktu eksplan biji berkecambah. Pada penelitian ini perlakuan eksplan biji yang terbaik adalah pada konsentrasi clorox 0% karena menunjukkan eksplan biji yang berkecambah lebih banyak. Lama perendaman clorox tidak memberikan pengaruh yang nyata terhadap inisiasi biji buah ajaib, namun mampu menurunkan persentase kontaminasi sebesar 83,33% dan persentase pencoklatan sebesar 100%.
