Abstract

This research aimed to determine the effect of different photoperiodes and various Plant Growth Regulator (PGR) of (2.4-D with BAP) on the callus induction of water apple leaves (Syzygium aqueum) var. Cincalo Weha in vitro. This research was conducted at the Plant Physiology and Biotechnology Laboratory, Sultan Ageng Tirtayasa University. This research used a randomized split-plot design with a completely randomized block design, 4 blocks as the repetition, and consisted of 2 factors. The first factor in the main plot was a photoperiod: 16 hours and 24 hours. The second factor as a sub-plot was various PGR of (2.4-D with BAP): without 2.4-D and BAP; 1 mg/l 2.4-D + 0.5 mg/l BAP; 2 mg/l 2.4-D + 0.5 mg/l BAP; and 3 mg/l 2.4-D + 0.5 mg/l BAP. The results showed that the treatment of photoperiod had no significant effect on callus diameter and callus area. While the treatment of various PGR of 2 mg/l 2.4-D + 0.5 mg/l BAP gave the best results on the parameters of callus diameter aged 8 Week After Planting (WAP) (1.05 cm) and 10 WAP (1.24 cm), as well as callus area aged 8 WAP (1.16 cm2) and 10 WAP (1.47 cm2). There was no interaction between the photoperiod and the PGR (2.4-D with BAP) in inducing callus. In this study, the texture of the callus was dominated by a friable texture and the color of the callus was dominated by yellow.

Additional Information: Jambu air var. Cincalo Weha merupakan plasma nutfah tanaman lokal yang berasal dari Desa Gondrong, Provinsi Banten. Jambu ini memiliki potensi ekonomi untuk dikembangkan, namun tidak diimbangi dengan ketersediaan bibit jambu air yang berkualitas. Kultur jaringan merupakan salah satu alternatif penyediaan bibit yang seragam, bebas penyakit dan dalam jumlah besar. Penelitian ini bertujuan untuk mengetahui efektivitas penyinaran dan ZPT (2,4-D dengan BAP) dalam menginduksi kalus daun jambu air var. Cincalo Weha secara in vitro. Rancangan percobaan yang digunakan Rancangan Petak Terbagi dengan Rancangan Acak Kelompok sebagai rancangan dasar, ulangan 4 blok dan terdiri dari 2 faktor. Faktor pertama adalah penyinaran: 16 jam (L1) dan 24 jam (L2). Faktor kedua adalah ZPT (2,4-D dan BAP): (tanpa 2,4-D dan BAP) (K0); (2,4-D 1 mg/L + BAP 0,5 mg/L) (K1); (2,4-D 2 mg/L + BAP 0,5 mg/L) (K2); dan (2,4-D 3 mg/L + BAP 0,5 mg/L) (K3). Hasil penelitian menunjukkan bahwa perlakuan penyinaran tidak berpengaruh terhadap diameter kalus dan luasan kalus. Sedangkan, perlakuan ZPT (2,4-D 2 mg/L + BAP 0,5 mg/L) memberikan hasil terbaik pada parameter diameter kalus umur 8 MST (1,05 cm) dan 10 MST (1,24 cm), serta luasan kalus umur 8 MST (1,16 cm2) dan 10 MST (1,47 cm2).
