Abstract

This study aimed to determine the genetic diversity in local ginger plants of Pandeglang Regency based on phenotype characteristics. genotype characteristics and the kinship relationships in local ginger plants of Pandeglang Regency based on phenotype and genotype characteristics. This research was conducted in Pandeglang Regency, Plant Biotechnology Physiology Laboratory, Faculty of Agriculture, University of Sultan Ageng Tirtayasa and BRIN LIPI Cibinong Genomics Building starting from August 2021 to March 2022. The results showed that based on the phenotype characteristics of the local ginger plant Pandeglang has a narrow genetic diversity (uniform). This can be known from the qualitative and quantitative character. From the results of dendogram analysis based on quantitative and qualitative characters, the closest kinship relationship is found in accessions B and D with coefficient values of 10, 303 while the farthest kinship is found in accessions A and C with coefficient values of 23, 835. Based on genotypic characteristics using ISSR markings the genetic diversity of local ginger plants has uniform diversity. This was because there was an equation in the number of bands that appear when analyzing using PCR. Thus based on the phenotype characteristics and genotype of the local ginger plant Pandeglang had a narrow (uniform) kinship relationship.

Penelitian ini dilakukan di Kabupaten Pandeglang, Laboratorium Fisiologi Bioteknologi Tanaman Fakultas Pertanian, Universitas Sultan Ageng Tirtayasa dan Gedung Genomik BRIN LIPI Cibinong yang dimulai pada bulan Agustus 2021 sampai dengan bulan Maret 2022. Hasil penelitian menunjukkan bahwa berdasarkan karakteristik fenotip tanaman jahe local Pandeglang memiliki keragaman genetic yang sempit (seragam). Hal ini dapat diketahui dari karakter kualitatif dan kuantitatif.. Dari hasil analisis dendogram berdasarkan karakter kuantitatif dan kualitatif hubungan kekerabatan terdekat teradapat pada aksesi B dan D dengan nilai koefisien 10, 303 sedangkan kekerabatan terjauh terdapat pada aksesi A dan C dengan nilai koefisien 23, 835. Berdasarkan karakteristik genotip menggunakan marka ISSR keragaman genetic tanaman jahe local memiliki keragaman yang seragam. Hal ini dikarenakan terdapat persamaan jumlah pita yang muncul saat analisis menggunakan PCR. Dengan demikian berdasarkan karakteristik fenotip dan genotip tanaman jahe local Pandeglang memiliki hubungan kekerabatan yang sempit (seragam).
