Abstract

This research aimed to determine the effect of IBA and kinetin levels on the shoots initiation of banana variety Mas Kirana (Musa acuminata Colla) in vitro. This research was conducted at the Integrated Agricultural System Network Cultivation Laboratory, Banten Province in March until October 2022. This research used a completely randomized design factorial. The first factor was the concentration of IBA which consists of 4 levels, spesifically 0, 1, 2, and 3 ppm. The second factor was the concentration of kinetin which consists of 4 levels, spesifically 0, 2, 4, and 6 ppm. Each treatment was repeated three times. The result showed that treatment of IBA 0 ppm gave the best results on the parameter of the time of shoots emergence with an average 5,80 days after planting, the number of shoots at 8 and 12 weeks after planting with an average 0,33 shoots, and shoots height at 8 weeks after planting with an average 0,43 cm. The concentration of kinetin had no significant effect on the parameters of the time of shoots emergence, the number of shoots, and the height of the shoots. In addition, there was no interaction between the two treatments.

Additional Information:	Pisang menjadi salah satu komoditas yang diminati oleh masyarakat. Pisang merupakan buah yang membuat perut kenyang sehingga produk olahan pisang bervariasi. Pisang Mas Kirana merupakan buah yang banyak dipilih oleh pengelola bisnis karena ukuran buah yang tidak terlalu besar maupun terlalu kecil. Selain itu, pisang Mas Kirana memiliki tekstur yang renyah serta rasanya yang manis dan segar. Pisang Mas Kirana mempunyai anakan sekitar 2-3 anakan per rumpun. Penerapan metode kultur jaringan dapat digunakan untuk meningkatkan produksi pisang. Metode kultur jaringan mampu menghasilkan individu baru yang seragam dengan waktu yang relatif singkat. Pemberian zat pengaruh tumbuh dapat membantu pertumbuhan tanaman. IBA dan kinetin dipilih menjadi zat pengatur tumbuh yang digunakan pada penelitian ini. Penelitian ini bertujuan untuk mengetahui pengaruh tingkat konsentrasi IBA dan kinetin terhadap pertumbuhan tunas pisang Mas Kirana secara in vitro. Penelitian ini dilaksanakan di Laboratorium Kultur Jaringan Sistem Pertanian Terpadu (SITANDU) Dinas Pertanian Provinsi Banten pada bulan Maret-Oktober 2022. Penelitian ini menggunakan Rancangan Acak Lengkap (RAL) faktorial. Faktor pertama adalah tingkat konsentrasi IBA dengan 4 taraf yaitu 0, 1, 2, dan 3 ppm. Faktor kedua adalah tingkat konsentrasi kinetin dengan 4 taraf yaitu 0, 2, 4, dan 6 ppm. Masing-masing perlakuan diulang sebanyak tiga kali. Hasil penelitian tanpa pemberian IBA memberikan hasil terbaik terhadap parameter waktu muncul tunas dengan rata-rata 5,80 HST, jumlah tunas umur 8 dan 12 MST dengan rata-rata 0,33 tunas serta tinggi tunas umur 8 MST dengan rata-rata 0,43 cm. Tingkat konsentrasi kinetin tidak berpengaruh terhadap parameter waktu muncul tunas, jumlah tunas, dan tinggi tunas. Selain itu, tidak terdapat interaksi antara pemberian IBA dan kinetin terhadap parameter waktu muncul tunas, jumlah tunas, dan tinggi tunas
