Abstract

Root endophytic fungi and arbuscular mycorrhizal (AMF) form a mutualistic symbiosis with their host plants. The purpose of this study was to determine the diversity of root endophytic fungi and AMF in Pteridophytes plants on Mount of Tukung Gede and their implications as a biology learning resource on Fungi concept. This study was conducted from July 2022 to January 2023. A random sampling method used for collected root endophytic fungi and AMF from three different station. Environmental parameters were also analyzed includes temperature, soil pH, soil moisture and light intensity. Root endophytic fungi and AMF were isolated by Pteridophytes roots then their could be identified, analyzed and calculated using by the Shannon index. Root endophytic fungi was isolated using PDA medium, while AMF fungi were isolated using pouring filter technique. The results showed that there were 8 genera of root endophytic fungi, namely Aspergillus, Trichoderma, Phoma, Phialophora, Penicillium, Paecilomyces, Humicola, and Fusarium, while 3 genera of AMF identified, namely Glomus, Acaulospora, and Gigaspora. The diversity index value of root endophytic fungi, which are Station I = 1.46 (medium), Station II = 1.28 (medium), and Station III = 1.33 (medium). The diversity of CMA, which are Station I = 0.69 (low), Station II = 0.59 (low), Station III = 0.41 (low). The results of this study can be utilized as a biology learning resource in the form of a booklet on fungi material.

Additional Information: Cendawan endofit akar dan mikoriza arbuskula (CMA) bersimbiosis mutualisme dengan tanaman inangnya. Penelitian ini bertujuan untuk mengetahui keanekaragaman cendawan endofit akar dan CMA pada tumbuhan Pteridophyta di Gunung Tukung Gede serta implikasinya sebagai sumber belajar biologi pada materi Fungi. Penelitian ini dilakukan pada Bulan Juli 2022 hingga Januari 2023. Pengambilan sampel dilakukan dengan metode random sampling pada tiga stasiun berbeda. Pengukuran parameter lingkungan meliputi suhu, pH tanah, kelembapan tanah dan intensitas cahaya. Cendawan endofit akar dan CMA diisolasi dari akar Pteridophyta lalu diidentikasi, dianalisis dan dihitung indeks Shannon-nya. Isolasi cendawan endofit akar menggunakan media PDA, sedangkan isolasi CMA dengan teknik tuang saring. Hasil penelitian menunjukkan cendawan endofit akar yang teridentifikasi terdapat 8 genus, yaitu Aspergillus, Trichoderma, Phoma, Phialophora, Penicillium, Paecilomyces, Humicola, dan Fusarium, sedangkan cendawan mikoriza arbuskula yang teridentifikasi terdapat 3 genus, yaitu Glomus, Acaulospora, dan Gigaspora. Nilai indeks keanekaragaman cendawan endofit akar pada, yaitu Stasiun I=1,46 (sedang), Stasiun II= 1,28 (sedang), dan Stasiun III= 1,33 (sedang). Adapun keanekaragaman CMA, yaitu Stasiun I= 0,69 (rendah), Stasiun II= 0,59 (rendah), Stasiun III= 0,41 (rendah). Hasil penelitian ini dapat dimanfaatkan sebagai sumber belajar biologi berupa booklet pada materi fungi.
