Dibimbing oleh Susiyanti, Imas Rohmawati, dan Juwartina Ida Royani. Indigofera merupakan tanaman dari kelompok kacang-kacangan (famili Fabaceae). Tanaman Indigofera telah banyak dimanfaatkan untuk pakan ternak dan konservasi tanah tetapi masih sangat terbatas dalam pengadaan benih dan bibitnya, sehingga diperlukan metode alternatif untuk perbanyakan tanaman Indigofera melalui kultur jaringan. Penelitian ini bertujuan untuk mendapatkan konsentrasi auksin 2,4-D dan sumber eksplan yang terbaik untuk inisiasi kalus tanaman Indigofera (Indigofera zollingeriana Miq.) dengan metode suspensi sel. Penelitian telah dilaksanakan di Laboratorium Pusat Teknologi Produksi Pertanian (PTPP) di Laboratoria Pengembangan Teknologi Industri Agro dan Biomedika (LAPTIAB), Badan Pengkajian dan Penerapan Teknologi (BPPT), kawasan PUSPIPTEK Tangerang Selatan. Rancangan yang digunakan adalah rancangan acak lengkap (RAL) faktorial, yang terdiri dari 2 faktor, yaitu auksin 2,4-D dan sumber eksplan. Faktor pertama adalah sumber eksplan yang terdiri dari 2 jenis, yaitu A1 (eksplan daun) dan A2 (eksplan batang). Faktor kedua adalah auksin 2,4- D yang terdiri dari beberapa taraf, yaitu B0 (MS), B1 (0,0 mg/L), B2 (1,0 mg/L), B3 (2,0 mg/L), B4 (3,0 mg/L), dan B5 (4,0 mg/L). Hasil penelitian inisiasi kalus Indigofera menunjukkan bahwa auksin 2,4-D sebanyak 2,0 mg/L dan 3,0 mg/L menghasilkan waktu muncul kalus tercepat (3,55 MST). Perlakuan tanpa auksin 2,4-D (hanya BAP) menghasilkan diameter kalus terbesar (1,21 cm). Penambahan auksin 2,4-D sebanyak 3,0 mg/L menghasilkan berat basah kalus terbesar (0,56 g). Kalus yang dihasilkan bertekstur remah dan berwarna putih krem. Eksplan batang memberikan waktu muncul kalus tercepat (3,51 MST). Eksplan daun memberikan diameter kalus terbesar (1,19 cm) dan berat basah kalus terbesar (0,52 g). Terdapat interaksi antara auksin 2,4-D dan sumber eksplan pada parameter diameter kalus, yaitu sebesar 1,54 cm (A1B1).

Guided by Susiyanti, Imas Rohmawati, and Juwartina Ida Royani. Indigofera was plant that used as feed and soil conservation. Propagation of Indigofera was still limited. Tissue culture was the alternative technique to propagated Indigofera for mass production. The aimed of this research was to callus initiation of Indigofera (Indigofera zollingeriana Miq.) through cell suspension culture using various concentration of 2,4-D and source of explants. These research was conducted at Laboratory of Center Agricultural Production Technology, LAPTIAB - BPPT from May to July 2018. The design experimental for this research was used completely randomized design (CRD) which arranged as factorial with two factors. The first factor was source of explants : A1 (leaf) and A2 (stem). The second factor was concentration of 2,4-D with several levels : B0 (MS), B1 (0.0 mg.L-1), B2 (1.0 mg.L-1), B3 (2.0 mg.L-1), B4 (3.0 mg.L-1), and B5 (4.0 mg.L-1). The results showed that the fastest of callus initiation on MS media added by 2,4-D with concentration 2.0 mg.L-1 and 3.0 mg.L-1 (3.55 WAP). The largest callus diameter showed on MS media without 2,4-D (1.21 cm of B1). The biggest wet weight of callus showed on MS media added by 2,4-D with concentration 3.0 mg.L-1 (0.56 g). Callus had friable textured and creamy white coloured. The fastest of callus initiation from source of explants was stem with 3.51 WAP meanwhile the leaf explants gave the largest callus diameter with 1.19 cm and the biggest wet weight of callus with 0.52 g. There was an interaction between 2,4-D and source of explants on callus diameter parameters (1.54 cm of A1B1). Keywords : 2,4-D auxin, callus initiation, explants, Indigofera plants
