Abstract
Both phosphate solubilizing bacteria and nitrogen fixing bacteria were known as Plant Growth Promoting Rhizobacteria. These bacteria have a capability to facilitate the absorption of nutrients such as phosphate (PO4) and nitrogen (NH3), that are available to plants. The aim of this study was to isolate and to characterize phosphate solubilizing bacteria and nitrogen-fixing bacteria from mangrove rhizosphere of Pulau Dua Santuary Serang-Banten. The bacteria samples were collected from 3 stations by purposive sampling technique. Samples of series dilution 10-3 up to 10-7 were plated on selective medium of Pikovskaya and NFB. The result showed that 5 genera of phosphate solubilizing bacteria and 2 genera of nitrogen-fixing bacteria have been isolated and characterized based on colony observation, gram staining, cell morphology, motility, and catalase test. The results of the study was then applied as learning module. The module was categorized as feasible (91%) as an alternative of learning source for X-grade of senior high school in the subconcept of Eubacteria. Additional Information: | ABSTRAK Bakteri pelarut fosfat dan bakteri penambat nitrogen asal rhizosfer dikenal sebagai bakteri pemacu pertumbuhan tanaman. Bakteri tersebut memiliki kemampuan memfasilitasi penyerapan nutrisi seperti fosfat (PO4) dan nitrogen (NH3), dan membuat nutrisi tersebut dalam bentuk yang tersedia bagi tanaman. Tujuan penelitian ini adalah untuk mengisolasi dan mengkarakterisasi bakteri pelarut fosfat dan bakteri penambat nitrogen asal rhizosfer mangrove Cagar Alam Pulau Dua Serang-Banten. Sampel bakteri dikoleksi dari 3 stasiun secara purposive sampling. Serial pengenceran 10-3-10-7 diinokulasikan pada media selektif Pikovskaya dan NFB. Hasil penelitian menunjukkan bahwa 5 genus bakteri pelarut fosfat dan 2 genus bakteri penambat nitrogen diisolasi dan dikarakteristik berdasarkan pengamatan koloni, pewarnaan gram, morfologi sel, motilitas, dan uji katalase. Hasil penelitian ini disajikan dalam bentuk modul pembelajaran. Modul ini dikategorikan sangat layak (91%) sebagai sumber belajar alternatif bagi siswa SMA kelas X pada subkonsep eubacteria.
