Abstract
Donna Carmen plant is one type of ornamental aglonema plant that is being loved during the pandemic. The attractiveness of aglonema plants is found in the shapes and color leaves. One way to obtain new plant genetic diversity is by mutation using the EMS (Ethyl Methane Sulphonate) mutagen. The aims of this study is to determine the effect of concentration and frequency of administration of Ethyl Methane Sulphonate on the success of mutations in aglonema cv. Donna Carmen (Aglaonema sp.) plant. The research was carried out from January to May 2021 at the Green House Jasa Alam Kaligandu, Serang City. This study used a factorial randomized block design (RAK) consisting of two factors. The first factor is the concentration of EMS (E) there are three levels, namely 100 ppm, 200 ppm, and 300 ppm. The second factor is the frequency of giving EMS (F) consisting of three levels, namely once every 2 days, every 4 days, and every 6 days and there is one control treatment as a comparison. So that there are 10 treatment combinations that were repeated 3 times, then 30 experimental units were obtained. Parameters observed were shoot height (cm), number of shoots (shoots), number of leaves (strands), leaf length (cm), leaf width (cm), leaf ratio, total chlorophyll (units), leaf area (cm2), shape leaves, leaf color, percentage of normal plants (%), percentage of mutant plants (%). The results showed that the concentration and frequency of EMS treatment had an effect on the success of mutations in the aglonema cv. Donna Carmen plant and there was no interaction between the two treatments. The administration of Ethyl Methane Sulphonate could inhibit the growth and development of plants compared to the control treatment and showed changes in the morphology of the aglonema cv. Additional Information: | Tanaman Donna Carmen merupakan salah satu jenis tanaman hias aglonema yang sedang digandrungi di masa pandemi. Daya tarik tanaman aglonema terdapat pada bentuk dan warna daunnya. Salah satu cara untuk mendapatkan keragaman genetik tanaman baru adalah dengan melakukan mutasi menggunakan mutagen EMS (Ethyl Methane Sulphonate). Penelitian ini bertujuan untuk mengetahui pengaruh konsentrasi dan frekuensi pemberian Ethyl Methane Sulphonate terhadap keberhasilan mutasi pada aglonema cv. tanaman Donna Carmen (Aglaonema sp.). Penelitian dilaksanakan pada bulan Januari hingga Mei 2021 di Green House Jasa Alam Kaligandu Kota Serang. Penelitian ini menggunakan Rancangan Acak Kelompok (RAK) faktorial yang terdiri dari dua faktor. Faktor pertama adalah konsentrasi EMS (E) ada tiga taraf yaitu 100 ppm, 200 ppm, dan 300 ppm. Faktor kedua adalah frekuensi pemberian EMS (F) yang terdiri dari tiga taraf yaitu 2 hari sekali, 4 hari sekali, dan 6 hari sekali serta terdapat satu perlakuan kontrol sebagai pembanding. Sehingga terdapat 10 kombinasi perlakuan yang diulang sebanyak 3 kali, maka diperoleh 30 satuan percobaan. Parameter yang diamati adalah tinggi pucuk (cm), jumlah pucuk (pucuk), jumlah daun (helai), panjang daun (cm), lebar daun (cm), rasio daun, klorofil total (satuan), luas daun (cm2), bentuk daun, warna daun, persentase tanaman normal (%), persentase tanaman mutan (%). Hasil penelitian menunjukkan bahwa konsentrasi dan frekuensi pemberian EMS berpengaruh terhadap keberhasilan mutasi pada aglonema cv. tanaman Donna Carmen dan tidak terjadi interaksi antara kedua perlakuan. Pemberian Ethyl Methane Sulphonate dapat menghambat pertumbuhan dan perkembangan tanaman dibandingkan dengan perlakuan kontrol dan menunjukkan perubahan morfologi aglonema cv. Pabrik Donna Carmen.
