Abstract

Rice is one of the staple food sources favored by most of the world's population, especially in developing countries such as Indonesia. In addition to its development in improving rice production and productivity, developments in improving the nutritional content of rice also need to be considered. One of the important micronutrients in rice that needs to be improved is very low folic acid. The occurrence of folic acid deficiency in the body is a problem that often occurs in developing countries, especially Indonesia which needs attention. Therefore, it is necessary to carry out molecular screening on putative transgenic rice cv Pare Gajah and cv Fatmawati which was successfully inserted folate gene to ensure the presence of these genes. This study aims to screen transgenic putative rice cv Fatmawati and cv Pare Gajah which was inserted with the folate gene was used as primer hpt and supported by a histochemical test with a hygromycin solution. This research was conducted from September to March 2022 at the Screen House of the Banten Agricultural Technology Research Center and the Biotechnology Laboratory, Faculty of Agriculture, Sultan Ageng Tirtayasa University. The research used descriptive quantitative and qualitative descriptive methods. The molecular research results using PCR produced and confirmed the presence of the hpt marker gene in putative transgenic rice using hpt primer. The putative transgenic rice plant cv Pare Gajah which has the hpt marker gene is found in PG6.1, PG6.2, PG6.3, PG6.5, PG6.6, PG6.8, PG6.9, and PG6.10. In putative cv Fatmawati transgenic rice, the hpt marker gene was confirmed in Ft3.2, Ft3.5, and Ft3.10. Histochemically, putative rice cv Pare Gajah transgenic which was resistant to hygromycin was found in PG6.1, PG6.3, PG6.5, and PG6.10. In putative cv Fatawati transgenic rice, plants that were resistant to hygromycin solution were found in Ft3.5 and Ft3.10. Additional Information: Padi merupakan salah satu sumber makanan pokok yang banyak digemari oleh sebagian penududuk di dunia, terutama negar-negara berkembang seperti Indonesia. Disamping pengembangannya dalam memperbaiki produksi dan produktivitas padi, pengembangan dalam memperbaiki kandungan nutrisi padi juga perlu diperhatikan. Salah satu mikronutrien penting pada beras yang perlu ditingkatkan adalah asam folat yang terkandung sangat rendah. Terjadinya kekurangan asam folat dalam tubuh merupakan masalah yang sering terjadi di negara berkembang khususnya Indonesia yang perlu mendapat perhatian. Oleh karena itu, perlu dilakukan skrining secara molekuler pada padi putatif transgenik cv Pare Gajah dan cv Fatmawati yang berhasil disisipkan gen folat untuk memastikan keberadaan gen tersebut. Penelitian ini bertujuan untuk menskrining padi putatif transgenik cv Fatmawati dan cv Pare Gajah yang disisipi gen folat dengan menggunakan primer hpt dan didukung dengan uji histokimia menggunakan larutan hygromisin. Penelitian ini dilaksanakan pada bulan September 2021 sampai Maret 2022 di Rumah Kaca Balai Penelitian Teknologi Pertanian Banten dan Laboratorium Bioteknologi Fakultas Pertanian Universitas Sultan Ageng Tirtayasa. Penelitian ini menggunakan metode deskriptif kuantitatif dan deskriptif kualitatif. Hasil penelitian molekuler secara PCR dengan primer hptt menghasilkan dan mengkonfirmasi keberadaan gen penanda hpt pada padi putatif transgenik. Tanaman padi transgenik putatif cv Pare Gajah yang memiliki gen penanda hpt secara molekuler terdapat pada PG6.1, PG6.2, PG6.3, PG6.5, PG6.6, PG6.8, PG6.9, dan PG6.10. Sedangkan pada padi putatif transgeni cv Fatmawati gen penanda hpt terkonfirmasi pada Ft3.2, Ft3.5, dan Ft3.10. Secara histokimia, padi putatif transgnik cv Pare Gajah yang tahan terhadap higromisin ditemukan pada PG6.1, PG6.3, PG6.5, dan PG6.10. Sedangkan pada padi putatif transgenik cv Fatawati, tanaman yang tahan terhadap larutan higromisin ditemukan pada Ft3.5 dan Ft3.10.
