Additional Information: Seaweed is a fishery commodity that has high economic value because of its wide utilization, both in daily life and in the industrial world. Types of Gracilaria sp. widely are used as raw material for making agar. One of the uses of agar is as a medium for microbial growth. The research aimed to determine the physical, chemical and biological characteristics of the agar media Gracilaria sp. based on the differences in pre-treatment concentrations according to the quality as standards for commercial agar media. The research was conducted in July - Oktober 2020 at Fishery Processing Products and Technology Laboratory of the Department of Fisheries Science, University of Sultan Ageng Tirtayasa. This research used three different concentration of NaOH (0%, 3%, 7%) asan alkaline pre-treatment in the agar media extraction process. The results showed, that the used of NaOH had an affected for on the quality of the agar media. The higher of NaOH concentration, increase the content of gel strength, ash content, moisture content, melting point, and general point while the sulfate content and yield value decreased. But reduce the yield significantly. Soaking NaOH solution with a concentration of 7% gets good results. Whereas in the Total Plate Count (TPC) test, it was found that the media with pre-treatment added 3% NaOH concentration was the best concentration that could grow as many bacteria as 2.27 x 10 2 cfu/mL. Agar media from Gracilaria sp. can be used as bacterial culture medium.
