Abstract

Mariam Rismawati, 2022, “Keragaman Genetik Plasma Nutfah Padi Lokal Indonesia Dan Introduksi Berbasis Marker Mikrosatelit Gen Umur Genjah Dan Ketahanan Wereng Batang Coklat Biotipe 3” Tesis. Program Pendidikan Magister Program Studi Ilmu Pertanian Pascasarjana Universitas Sultan Ageng Tirtayasa. Pembimbing I Dr. Susiyanti, S.P., M.P, Pembimbing II Dr. Zahratul Millah, S.P, M.Si. Kata Kunci : Oryza sativa L, Plasma Nutfah, Keragaman Genetik, SSR Padi (Oryza sativa L.) merupakan tanaman pangan penting yang di tanam hampir sepertiga dari jumlah total bahan pangan di dunia. Plasma nutfah padi berupa varietas lokal yang memiliki keunggulan genetik secara turun temurun. Penggunaan varietas padi berumur genjah dan tahan wereng batang coklat biotipe 3 akan menguntungkan dalam banyak hal, diantaranya adalah mengurangi resiko gangguan lingkungan hama, penyakit, dan gagal panen. Penelitian ini bertujuan untuk Mengeksplorasi padi lokal Indonesia dan Introduksi yang memiliki sifat umur genjah dan tahan terhadap hama wereng batang coklat biotipe 3 berdasarkan keragaman genetik. Penelitian ini menggunakan metode clustering yang digunakan untuk membangun pohon filogeneti dan selanjutnya menganalisis kekerabatan dan elektroforegram. Hasil penelitian menunjukan Keragaman Genetik aksesi plasma nutfah padi yang memiliki umur genjah dan tahan wereng batang coklat biotipe 3 berbasis marka SSR memiliki kemiripan 87-100% atau jarak genetiknya yaitu 0-13% yaitu pada kelompok I sub kelompok A dan B. Penggunaan primer SSR (RM6838, RM5607 dan RM17) menghasilkan pola pita polimorfis yang memiliki nilai Polymorphic Information Content (PIC) ≥0,5 yang dapat digunakan sebagai alat marker penseleksi gen umur genjah dan gen tahan wereng batang coklat biotipe 3.

Rice germplasm in the form of local varieties that have genetic advantages from generation to generation. The use of early maturing and resistant brown planthopper biotype 3 rice varieties will be beneficial in many ways, including reducing the risk of environmental disturbances, pests, diseases, and crop failure. This study aims to explore Indonesian local rice and introductions that have early maturity traits and are resistant to biotype 3 brown planthopper pests based on genetic diversity. This study uses the clustering method which is used to build a phylogenetic tree and then analyzes the relationship and electrophorogram. The results showed that the genetic diversity of rice germplasm accessions with early maturity and resistance to brown planthopper biotype 3 based on SSR markers had a similarity of 87- 100% or genetic distance of 0-13%, namely in group I, subgroups A and B. Primary use of SSR (RM6838, RM5607 and RM17) produced a polymorphic banding pattern that had a Polymorphic Information Content (PIC) value of 0.5 which could be used as a marker tool for selecting early maturing genes and brown planthopper biotype 3 resistant genes.
